摘 要:耐草甘膦和耐草铵膦是转基因作物育种重要的目标性状。将耐草甘膦基因MC1-EPSPS构建到含有bar基因的植物表达载体p TF101.1中,通过农杆菌介导法转入玉米材料Hi-II中,从而获得兼具耐受草甘膦和草铵膦性状的转基因玉米材料CM8401。目的基因PCR检测显示,MC1-EPSPS和bar基因稳定整合到玉米基因组中。目的蛋白试纸条检测结果显示,MC1-EPSPS蛋白和PAT蛋白在转基因玉米世代间中表达稳定。田间除草剂耐受性鉴定试验表明,转基因玉米CM8401对草甘膦和草铵膦都具有良好耐受性,可耐受4倍推荐中剂量的草甘膦和草铵膦。Glyphosate resistance and glufosinate resistance are important target traits in genetically modified crop breeding.Glyphosate resistance gene MCI-EPSPS was constructed into the plant expression vector pTF101.1 containing bar gene in the present study.The transgenic maize material CM8401 with tolerance to glyphosate and glufosinate was obtained by Agrobacterium mediated transformation into maize material Hi-II.PCR detection of target genes showed that MCI-EPSPS and bar genes were statically integrated into maize genome,and the test results of target protein strip showed that MC1-EPSPS and PAT proteins expressed statically in transgenic maize between generations.Field herbicide tolerance identification tests showed that the transgenic maize CM8401 exhibited good tolerance to both glyphosate and glufosinate,and could tolerate 4 times the recommended medium dose.
