摘  要:将48只体质量为(20±2)g的雄性昆明小鼠随机分为空白对照组、模型组、凯西莱阳性对照组及异甘草素(isoliquiritigenin,ISL)低剂量组、ISL中剂量组和ISL高剂量组等6个组。对小鼠进行酒精灌胃30 d,建立酒精性肝损伤模型。利用凯西莱及低、中、高剂量ISL治疗10 d,记录小鼠体质量、采食量,肝脏测质量计算肝脏系数。检测小鼠血清中的谷丙转氨酶(ALT)、谷草转氨酶(AST)活力、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、丙二醛(MDA)、谷胱甘肽(GSH)和超氧化物歧化酶(SOD)含量。同时,分别用qRT-PCR和ELISA检测炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)在肝脏组织中的mRNA和蛋白表达量,并观察肝脏的病理组织切片。结果显示,成功构建了肝损伤模型,与模型组相比,ISL各组小鼠肝小叶和肝细胞形态结构有所恢复,脂肪空泡减少,相关指标得到改善。一般生理指标检测中,ISL各组小鼠体质量和采食量均有所恢复,肝脏系数极显著降低(P<0.01)。血清肝功能指标检测中,ISL中剂量组AST活性显著性降低(P<0.05),ISL各组TC、TG和LDL含量显著性降低(P<0.05),ISL高剂量组HDL含量显著增高(P<0.05)。氧化应激指标检测中,ISL各剂量组GSH和SOD含量升高,ISL低剂量组MDA含量显著性降低(P<0.05)。在炎症指标的检测中,ISL各剂量组IL-1β和TNF-α mRNA含量极显著降低(P<0.01),ISL低、中剂量组IL-6 mRNA含量显著降低(P<0.05)。ISL中、高剂量组IL-1β蛋白含量显著下降(P<0.05),ISL各剂量组IL-6蛋白含量极显著下降(P<0.01),ISL高剂量组TNF-α蛋白含量下降。结果表明,ISL对酒精性肝损伤小鼠肝脏有一定的保护作用。To investigate the protective effect of isoliquiritigenin(ISL) on alcoholic liver disease in mice,forty-eight male Kunming mice were randomly divided into six groups:the blank control group,model group,positive control group,low-dose group,medium-dose group and the high-dose group.The alcoholic liver disease model was established by gavage with alcohol for 30 d.The mice were treated with various doses of ISL for 10 d.The body weight and feed intake were recorded,and the liver was weighted to calculate the liver index.The contents of ALT,AST,TC,TG,HDL,LDL,MDA,GSH and SOD in serum of mice were detected.The mRNA and protein expressions of IL-1β,IL-6 and TNF-α in liver were measured by qRT-PCR and ELISA,and the pathological sections of liver were observed by light microscopy.The alcoholic liver disease model was successfully constructed.Compared with the model group,the morphology and structure of liver lobules and hepatocytes in the whole ISL groups were restored,fat vacuoles were reduced,and relevant indicators were ameliorated.In the detection of general physiological indexes,the body weight and feed intake of mice in the whole ISL groups were recovered,and the liver index was extremely significantly decreased(P <0.01).The results of the detection of serum liver function indicators showed that AST activity was significantly decreased in the medium-dose group of ISL(P<0.05),TC,TG and LDL contents were significantly decreased in the whole ISL groups of ISL(P<0.05),and HDL content was significantly increased in the high-dose group of ISL(P<0.05).The results of the detection of oxidative stress indexes showed that the contents of GSH and SOD increased in the whole ISL groups,while the content of MDA decreased significantly in the low-dose group of ISL(P<0.05).In the detection of inflammatory indicators results showed that IL-1βand TNF-αmRNA contents were extremely significantly reduced in the whole ISL groups(P<0.01),and IL-6mRNA content was significantly reduced in the low-and medium-dose groups of ISL(P<0.05).IL
