摘  要:将新采集且活力达标的湖羊精液添加到含不同质量浓度(0.00,0.25,0.50,1.00,2.00 mg/L,Ⅰ~Ⅴ组)纳米颗粒硒(nanoparticulate selenium,SeNPs)的冷冻基础稀释液中进行稀释,然后将稀释的精液分装入0.25 mL冻精细管,并在液氮中储存。7 d后解冻,对每个处理的精液进行精子质量参数评估,包括精子活力、运动参数、畸形率、顶体完整率、质膜完整性,并测定各组精液的总抗氧化能力(T-AOC)、过氧化氢酶活性(CAT)、活性氧(ROS)和丙二醛(MDA)含量以及相关抗氧化基因(SOD1、GCLC、GCLM和PRDX1)的表达。结果显示,与Ⅰ组相比,Ⅲ,Ⅳ组对解冻后精子的运动能力、顶体完整率和膜完整性均有积极影响(P<0.05),尤其是Ⅳ组精子冻后活力达48.68%,顶体完整率为80.51%,头部质膜完整率为52.54%,尾部质膜完整率为50.99%,均处于最高水平;SeNPs处理后的冷冻精液T-AOC和CAT活性显著增加(P<0.05),其中Ⅳ组T-AOC最高为(10.94±0.25) U/mL、CAT活性为(11.01±0.92)U/mL。SeNPs显著降低了解冻后精子内MDA和ROS水平且以1.0 mg/L质量浓度处理最佳(P<0.05),MDA含量可降至(4.81±0.36)mmol/L,ROS水平为1 880.98±44.86。抗氧化基因检测显示,1.00 mg/L SeNPs处理组SOD1极显著高于0.00 mg/L SeNPs处理组(P<0.01),GCLM、GCLC和PRDX1显著高于0.00 mg/L SeNPs处理组(P<0.05)。结果表明,在湖羊冷冻稀释液内SeNPs质量浓度为1.00 mg/L时,可以使精子结构和功能整体得到改善,而且对冻融精子抗氧化能力也有积极作用。The newly collected and viability semen of lake sheep was added to the frozen basal dilution containing nanoparticulate selenium(SeNPs) with different mass concentrations(0.00,0.25,0.50,1.00,2.00 mg/L,groups Ⅰ-Ⅴ) for dilution,and then the diluted semen was divided into 0.25 mL frozen fine tubes and stored in liquid nitrogen.After 7 d of thawing,sperm quality parameters were evaluated for each treated semen,including sperm motility,motility parameters,malformation rate,acrosome integrity rate,plasma membrane integrity,and the total antioxidant capacity(T-AOC),catalase activity(CAT),reactive oxygen species(ROS) and malondialdehyde(MDA) content of each group of semen and the expression of related antioxidant genes(SOD1,GCLC,GCLM and PRDX1) were determined.The results showed that compared with group Ⅰ,groups Ⅲ and Ⅳ had positive effects on sperm motility,acrosome integrity rate and membrane integrity after thawing(P<0.05),especially the post-freezing motility of sperm in groupⅣreached 48.68%,the acrosome integrity rate was 80.51%,the plasma membrane integrity rate of the head was 52.54%,and the integrity rate of the caudal plasma membrane was 50.99%,all at the highest level.The TAOC and CAT activities of frozen semen treated with SeNPs increased significantly(P<0.05),and the highest T-AOC in group Ⅳ was(10.94±0.25)U/mL,and CAT activity was(11.01±0.92)U/mL.SeNPs significantly reduced MDA and ROS levels in spermatozoa after defrost and were best treated at a mass concentration of 1.0 mg/L(P<0.05),MDA content could be reduced to(4.81±0.36)mmol/L,and ROS levels were 1 880.98±44.86.The antioxidant gene assay showed that SOD1was extremely significantly higher in the 1.00mg/L SeNPs-treated group than in the0.00mg/L SeNPs-treated group(P <0.01),and GCLM,GCLC and PRDX1 were significantly higher than in the 0.00 mg/L SeNPs-treated group(P <0.05).The results indicated that the SeNPs mass concentration of 1.00mg/L within the frozen dilution solution of lake sheep resulted in an overall improvement in sperm stru
