摘 要:目的探讨脂肪酰胺水解酶(fatty acid amide hydrolase,FAAH)385C/A多态性位点与中国汉族人群阿片类药物依赖的相关性。方法选取汉族人群阿片类药物依赖者308例(实验组)和健康人群631例(对照组)。采用限制性片段长度多态性聚合酶链反应(restriction fragment length polymorphism polymerase chain reaction,RFLP-PCR)和基因测序对FAAH 385C/A多态性位点进行分型。采用SPSS 13.0进行统计分析。结果实验组携带A等位基因频率显著高于对照组(P=0.002,OR=1.34,95%CI=1.11-1.62)。同质和异质遗传模式分析发现实验组AA、AC基因型频率均显著高于对照组(P<0.05)。调整性别、年龄等混杂因素后结果相似(同质:P=0.041,OR=2.30,95%CI=1.04-5.12;异质:P=0.004,OR=1.55,95%CI=1.16-2.08)。实验组按同质(AA vs.CC)和异质遗传模式(AC vs.CC)分层分析,性别、年龄、BMI与阿片类药物依赖均无统计学差异(P>0.05)。结论FAAH基因385C/A多态性位点可能是汉族阿片类药物依赖的遗传易感多态性位点。Objective To explore the association between 385C/A polymorphism of fatty acid amide hydrolase(FAAH)and opioid dependence.Methods A total of 308 opioid dependent patients(cases)and 631 healthy subjects(controls)were recruited from the Compulsory Detoxification Center of Nanchong and Affiliated Hospital of North-Sichuan Medical College,respectively.Restriction fragment length polymorphism polymerase chain reaction(RFLP-PCR)and direct sequencing genotyping were performed to detect the presence of the FAAH 385C/A genetic polymorphism.SPSS for Windows software package version 13.0 were applied for statistical analysis.Results The frequency of A allele of FAAH 385C/A polymorphism in the cases was significantly higher compared with that in the controls(P=0.002,OR=1.34,95%CI=1.11-1.62).The frequency of AA and AC genotypes of FAAH 385C/A polymorphism in the cases were higher than those in controls(P<0.05).Similar results were obtained after adjusting for age and sex(AA us.CC:P=0.04,OR=2.30,95%CI=1.04-5.12;AC us.CC:P=0.004,0R=1.55,95%CI=1.16-2.08).Additionally,stratified analysis by AA and AC genotype showed no difference in gender,age,and body mass index(BMI)between two groups(P>0.05).Conclusion The FAAH 385C/A polymorphism might contribute to opioid dependence in Chinese Han population.
