过敏性鼻炎患儿血清miR-124-3p和GATA3表达水平与疾病严重程度、Th1/Th2平衡的关系

摘　　要：目的探讨过敏性鼻炎(AR)患儿血清微RNA-124-3p(miR-124-3p)、GATA结合蛋白3(GATA3)表达与患儿疾病严重程度、Ⅰ型辅助性T细胞(Th1)/Ⅱ型辅助性T细胞(Th2)平衡的相关性。方法选择2020年6月至2022年4月在江苏省无锡市妇幼保健院(以下简称“我院”)儿童保健耳鼻喉科确诊的AR患儿87例为观察对象(AR组),按症状严重程度分为轻度组53例、中重度组34例,另选取同期在我院健康体检儿童85例为对照组。采用实时荧光定量PCR(qRT-PCR)法检测血清miR-124-3p表达水平,流式细胞术检测Th1、Th2细胞比例及Th1/Th2,采用酶联免疫吸附法检测血清γ干扰素(IFN-γ)、白细胞介素12(IL-12)、IL-4、IL-5、IL-10、GATA3水平,Pearson法分析AR患儿血清miR-124-3p表达与GATA3相关性,mi R-124-3p、GATA3与Th1/Th2及细胞因子相关性。结果AR组血清mi R-124-3p表达水平低于对照组,GATA3水平高于对照组,差异有统计学意义(P<0.05);中重度组血清mi R-124-3p表达水平低于轻度组,GATA3水平高于轻度组,差异有统计学意义(P<0.05)。AR组Th1细胞比例和Th1/Th2比值低于对照组,Th2细胞比例高于对照组,差异有统计学意义(P<0.05)。AR组血清IFN-γ、IL-12水平低于对照组,IL-4、IL-5、IL-10水平高于对照组,差异有统计学意义(P<0.05)。相关性分析显示,AR患儿血清miR-124-3p表达水平与GATA3水平呈负相关(r<0,P<0.05)。miR-124-3p与Th1、Th1/Th2、IFN-γ、IL-12呈正相关(r>0,P<0.05),与Th2、IL-4、IL-5、IL-10呈负相关(r<0,P<0.05);GATA3与Th1、Th1/Th2、IFN-γ、IL-12呈负相关(r<0,P<0.05),与Th2、IL-4、IL-5、IL-10呈正相关(r>0,P<0.05)。结论AR患儿血清mi R-124-3p表达水平降低,GATA3水平升高,与患儿疾病严重程度和Th1/Th2失衡有关。Objective To investigate the correlation between the expression of serum microRNA-124-3p(miR-124-3p)and GATA binding protein 3(GATA3)and the severity of the disease and the balance of typeⅠhelper T cells(Th1)/typeⅡhelper T cells(Th2)in children with allergic rhinitis(AR).Methods A total of 87 children with AR diagnosed in the Department of Otolaryngology Children’s Health Care of Wuxi Maternal and Child Health Care Hospital(hereinafter referred to as“our hospital”)from June 2020 to April 2022 were selected as the observation objects(AR group),and were divided into mild group(53 cases),moderate and severe group(34 cases)according to the severity of symptoms,and 85 healthy children undergoing physical examination in our hospital during the same period were selected as the control group.Real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the expression level of serum miR-124-3p,and flow cytometry was used to detect the proportion of Th1,Th2 cells,and Th1/Th2.Serum levels of interferon-γ(IFN-γ),interleukin-12(IL-12),IL-4,IL-5,IL-10 and GATA3 were detected by enzyme-linked immunosorbent assay.The correlation between the expression of miR-124-3p and GATA3 in children with AR was analyzed by Pearson method,while miR-124-3p and GATA3 were associated with Th1/Th2 and cytokines.Results The expression level of miR-124-3p in AR group was lower than that in control group,while the level of GATA3 was higher than that in control group,and the differences were statistically significant(P<0.05).The expression level of miR-124-3p in the moderate and severe group were lower than that in the mild group,while the level of GATA3 was higher than that in the mild group,and the differences were statistically significant(P<0.05).The proportion of Th1 cells and Th1/Th2 ratio in AR group were lower than that in control group,while the proportion of Th2 cells was higher than that in control group,and the differences were statistically significant(P<0.05).The serum levels of IFN-γand IL-12 in AR group were lower th
