摘 要:目的探讨蒺藜生药粉对大鼠的长期毒性。方法将120只SD大鼠随机分为对照组(等体积饮用水)及低、中、高剂量组(蒺藜生药粉2.13,4.25,8.50 g/kg),各30只。各组大鼠灌胃给予相应药物或等体积饮用水(15 mL/kg),每天1次,连续30 d。观察末次给药后及末次给药30 d后大鼠的体质量、一般情况(形态外观、中毒反应症状等);采用苏木素-伊红染色,观察肝脏组织病理学变化情况;检测血液细胞、血液生化、血清电解质、凝血指标变化情况。结果给药期间各给药组大鼠均未见死亡及明显毒性反应。开始给药至末次给药30 d后各给药组大鼠体质量均呈自然增长趋势,末次给药后及末次给药30 d后与对照组比较无明显差异(P>0.05)。末次给药后,各给药组出现不同程度灶状炎性细胞浸润,且高剂量组可见肝细胞脂肪变性,末次给药30 d后均有不同程度改善。末次给药后与对照组比较,中剂量组雌性大鼠碱性磷酸酶(ALP)水平明显降低(P<0.05);低剂量组雄性大鼠红细胞计数(RBC)、血红蛋白(Hb)水平、平均红细胞血红蛋白浓度明显升高,平均红细胞体积(MCV)、平均红细胞血红蛋白量(MCH)、网织红细胞水平明显降低,中剂量组雄性大鼠MCH明显降低,高剂量组雄性大鼠RBC及Hb水平明显升高(P<0.05)。末次给药30 d后与对照组比较,高剂量组雌性大鼠MCV明显升高,低、中、高剂量组雌性大鼠ALP及中剂量组雌性大鼠γ-谷氨酰转肽酶水平明显降低(P<0.05);低、高剂量组雄性大鼠淋巴细胞百分比明显升高(P<0.05)。结论长期(≥30 d)给予高剂量(≥8.50 g/kg)蒺藜生药粉对大鼠具有一定的肝毒性,但随停药时间的延长大鼠可自行恢复。Objective To investigate the long-term toxicity of Tribuli Fructus crude powder on rats.Methods A total of 120 SD rats were selected and randomly divided into the control group(equal volume of drinking water),the low-,medium-,and high-dose groups(2.13,4.25,8.50 g/kg of Tribuli Fructus crude powder),with 30 rats in each group.Each group was given corresponding drugs or equal volume of drinking water(15 mL/kg)by gavage,once a day for 30 d.The body weight and general condition(appearance,toxic reaction symptoms and so on)in each group after the last administration and 30 d after the last administration was determined or observed.Hematoxylin-eosin staining method was used to observe pathological changes in liver tissue.The changes in indicators related to blood cells,blood biochemistry,serum electrolytes and coagulation were detected.Results During the administration,no death or obvious toxic reaction was observed in the low-,medium-and high-dose groups.The body weight in the low-,medium-and high-dose groups showed a normal growth from the start to 30 d after the last administration,and that was similar to that in the control group after the last administration and 30 d after the last administration(P>0.05).After the last administration,different degrees of focal inflammatory cell infiltration were observed in the low-,medium-and high-dose groups,and hepatocellular steatosis was observed in the high-dose group,while the above symptoms in the above groups improved to varying degrees 30 d after the last administration.After the last administration,compared with those in the control group,the alkaline phosphatase(ALP)level of female rats in the medium-dose group was significantly lower(P<0.05);the red blood cell count(RBC),hemoglobin(Hb)level and mean corpuscular hemoglobin concentration(MCHC)of male rats in the low-dose group were significantly higher,the mean corpuscular volume(MCV),mean corpuscular hemoglobin(MCH)and reticulocyte(RET)level were significantly lower;the MCH of male rats in the medium-dose group was sig
