摘 要:目的探讨补肾通络方治疗膝骨性关节炎可能的作用机制。方法通过BATMAN-TCM数据库筛选补肾通络方的活性成分及潜在靶点。通过CTD数据库、人类孟德尔遗传数据库、GeneCards数据库检索膝骨性关节炎相关基因,并通过CoolGeN数据库筛选;将制剂潜在靶点与疾病相关基因相互映射,得补肾通络方作用于膝骨性关节炎的靶点基因,导入String数据库进行网络拓扑属性分析,筛选重要靶点蛋白。将补肾通络方作用于膝骨性关节炎的靶点基因导入DAVID数据库分别进行基因本体论(GO)功能富集分析及京都基因与基因组百科全书(KEGC)信号通路富集分析。结果筛选出70个活性成分,其中熟地2个、杜仲11个、当归16个、山萸肉2个、怀牛膝3个、寄生5个、独活9个、秦艽3个、防风4个、细辛6个、地龙3个、甘草16个,10个成分为多种中药共有。共得到46个共有靶点,其中重要蛋白靶点10个。共富集到GO条目133个,包括生物过程96个、细胞组分14个、分子功能23个,涉及细胞凋亡过程、胞质溶胶、乙酰胆碱结合等;KEGG信号通路76条,包括肿瘤坏死因子(TNF)信号通路、丝裂原活化蛋白激酶(MAPK)信号通路、破骨细胞分化通路等。结论补肾通络方可能通过多活性成分、多靶点、多通路治疗膝骨性关节炎。Objective To investigate the mechanism of Bushen Tongluo Recipe in the treatment of knee osteoarthritis.Methods The active ingredients and potential targets of Bushen Tongluo Recipe were screened by the BATMAN-TCM database.The knee osteoarthritis-related genes were searched by the Comparative Toxicogenomics Database,Online Mendelian Inheritance in Man and GeneCards and screened by the CooLGeN database.The potential targets of the preparation were intersected with disease-related genes to obtain the target genes for the treatment of knee osteoarthritis with Bushen Tongluo Recipe,and the above target genes were imported into the String database for network topology attribute analysis to screen the important target proteins.The above target genes were imported into the DAVID database for gene ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway enrichment analysis.Results Seventy active ingredients were obtained,two from Rehmanniae Radix Praeparata,eleven from Eucommiae Cortex,sixteen from Angelicae Sinensis Radix,two from Corni Fructus,three from Achyranthis Bidentatae Radix,five from Visci Herba,nine from Angelicae Pubescentis Radix,three from Gentianae Macrophyllae Radix,four from Saposhnikoviae Radix,six from Asari Radix et Rhizoma,three from Pheretima,and sixteen from Glycyrrhizae Radix et Rhizoma.Among the above ingredients,ten were common to various traditional Chinese medicines.A total of 46 common targets were obtained,including 10 important protein targets.A total of 133 GO entries were collected,including 96 biological processes,14 cellular components,and 23 molecular functions,involving cell apoptosis process,cytosol,acetylcholine binding and so on.A total of 76 KEGG signaling pathways were obtained,including tumor necrosis factor(TNF),mitogen-activated protein kinase(MAPK),osteoclast differentiation and other signaling pathways.Conclusion Bushen Tongluo Recipe may treat knee osteoarthritis through multiple active ingredients,targets and pathways.
