摘  要:目的研究维生素D受体(VDR)在小鼠子宫内膜中的表达及其对胚胎着床的影响。方法免疫组化及Western Blot法检测孕1~6 d小鼠子宫内膜VDR表达;孕2 d小鼠一侧子宫角注入25μl 1 mM Calcifediol(VDR抑制剂)处理,另一侧子宫角注入等体积溶剂对照,孕4 d收获小鼠子宫组织,检测子宫内膜接受态标志分子白血病抑制因子(LIF)和整合素ανβ3的表达,或孕6 d观察胚胎着床情况。结果VDR在孕1~5 d小鼠子宫内膜中的表达呈现动态变化,孕1~2 d主要在上皮表达,基质表达较弱,孕3~4 d开始基质表达逐渐增强,孕5 d达到高峰,孕6 d着床区VDR表达显著高于非着床区(P<0.01);与对照侧比较,VDR抑制剂Calcifediol处理组孕4 d小鼠子宫内膜LIF和整合素ανβ3蛋白的表达均显著下降(P<0.05),孕6 d胚胎着床数亦明显下降(P﹤0.05)。结论VDR在孕1~6 d小鼠子宫内膜中呈动态表达,着床区VDR表达显著高于非着床区,而抑制子宫内膜VDR可降低子宫接受态使胚胎着床受损。Objective To investigate the expression of vitamin D receptor(VDR)in mouse endometrium and its role in embryonic implantation.Methods The expression of VDR on pregnant days 1~6 was examined by immunohistochemistry(IHC)and Western Blot.On pregnant day 2,the VDR inhibitor Calcifediol(25μl,1 mM)was directly injected into one side of the pregnant mouse uteri and the other side was treated with the same amount of solvent as control.Then,the expressions of leukocyte inhibitory factor(LIF)and integrinανβ3 were examined on pregnant day 4,and implantation sites were observed on pregnant day 6.Results Results of IHC showed that VDR exhibited a spatiotemporal expression manner in mouse uteri and results of Western Blot revealed that the abundance of VDR protein in mouse uteri was gradually increased on pregnant days 1~5,with a peak level on pregnant day 5.Moreover,on pregnant day 6,VDR expression in implantation site was significantly higher than that in non-implantation site(P<0.01).Compared with the control side,on pregnant day 4,the injection of Calcifediol into one side of mouse uterine horns resulted in impaired endometrial receptivity by inhibiting the expressions of LIF and integrinανβ3(P<0.05),finally affected implantation on day 6(P<0.05).Conclusion VDR involves in the establishment of the endometrial receptivity,which affects embryonic implantation.
