摘 要:【目的】探究生长调节剂NAA+IBA对圆齿野鸦椿插穗生根及其氧化酶(POD、PPO、IAAO)活性和内源激素(IAA、ABA、GA3、ZR)含量的影响,初步揭示圆齿野鸦椿扦插生根的生化机理,为促进圆齿野鸦椿扦插生根提供理论依据。【方法】于2—3月圆齿野鸦椿腋芽开始萌动时剪取1年生的成熟枝条制备插穗,并分成两组进行处理。处理I:插穗基部在150 mg/LNAA+150 mg/LIBA的混合溶液中浸泡2 h;处理II:插穗基部置清水中浸泡2 h作为对照(CK)。处理结束后立即进行取样,扦插后每隔15 d取样1次,观察记载其生根过程的形态变化,并测定不同取样时期插穗基部长2 cm的皮层内氧化酶活性与内源激素含量。【结果】圆齿野鸦椿插穗以愈伤组织生根型为主。处理I与处理II相比,愈伤组织形成时间可提早15 d左右,不定根的形成时间可提早30 d左右。扦插第0天,处理I和处理II相比,插穗POD和PPO活性显著升高,而IAAO活性显著降低;IAA、GA3和ABA含量显著升高,ZR含量则无明显变化。在整个生根过程中,两个处理插穗POD和PPO活性的变化趋势一致,均表现为“先升后降”,且处理I在扦插后第30天达到峰值,而处理II的峰值出现在扦插后第45天;在整个生根过程中,IAA含量处理I均显著高于处理II。【结论】圆齿野鸦椿属扦插生根较困难树种。插穗经150 mg/L NAA+150 mg/L IBA混合溶液浸泡处理2 h可加快其生根进程。提高插穗皮层内POD和PPO活性,降低IAAO活性,以及提高IAA含量及其与ABA、GA3、ZR的比值,有利于促进圆齿野鸦椿插穗愈伤组织和不定根的形成。【Objective】To explore the effects of exogenous hormones on rooting and the activities of oxidase(POD,PPO,IAAO)and the contents of endogenous hormones(IAA,ABA,GA3,ZR)of cuttings of Euscaphis konishii,and preliminally reveal the biochemical mechanism of cutting rooting of E.konishii,and provide theoretical basis for promoting cutting rooting of E.konishii.【Method】From February to March in early spring,when the axillary buds began to sprout,the mature branches of the previous year were cut to prepare cuttings and divided into two groups for treatment.Treatment I:the base of cuttings was soaked for 2 h in an equal mixture of 150 mg/L NAA+150 mg/L IBA;Treatment II:the cuttings were immersed in clean water for 2 hours as control(CK).Samples were taken immediately after the treatment,and samples were taken every 15 days after cutting.Morphological changes during rooting were observed,and oxidase activities and endogenous hormone contents in the cortex with a length of 2 cm of the cuttings at different sampling periods were measured.【Result】Callus rooting was the main type of cuttings.Compared with treatment II,callus formation time of treatment I was about 15 d earlier and adventitious root formation time was about 30 d earlier.On the 0th day of cuttings,compare with treatment I and II,the activities of POD and PPO were significantly increased,but the activities of IAAO were significantly decreased.The contents of IAA,GA3 and ABA increased significantly,while the contents of ZR had no significant change.During the whole rooting process,the change trend of POD and PPO activities of the two treatments was the same,showing“first rise and then fall”,and the peak value of treatment I was reached on the 30th day after cutting,while that of treatment II was reached on the 45th day after cutting.IAA content in treatment I was significantly higher than that in treatment II during the whole rooting process.【Conclusion】E.konishii is difficult to take rooting cuttings.The rooting process of cuttings is accelerat
