摘  要:为筛选快速、有效的花粉活力测定方法,研究花粉特性,以本地优良品种圆核桃的花粉为材料,以失活花粉为对照,分别采用醋酸洋红染色法、氯化三苯基四氮唑(TTC)法、MTT染色法、碘-碘化钾(I2-KI)染色法测定其花粉活力。结果表明,MTT染色法可快速有效地检测本地圆核桃花粉活力;本地圆核桃花粉可选用冷藏保存,在冷藏保存40 d后,花粉活力还维持在较高水平。本试验中,圆核桃花粉进行杂交时,不能采用泡水喷施的方法;其雄性花序在自然散粉16 d后失活。In order to screen a rapid and effective method for determining pollen viability and study pollen characteristics,the pollen of local excellent variety Juglans regia was used as the material,and the inactivated pollen was used as the control.The pollen viability was determined by acetocarmine staining,triphenyltetrazolium chloride(TTC)staining,MTT staining and iodine-potassium iodide(I 2-KI)staining.The results showed that MTT staining method could quickly and effectively detect the pollen viability of local round walnut.The local round walnut pollen could be stored in cold storage.After 40 days of cold storage,the pollen viability remained at a high level.In this experiment,the method of soaking and spraying could not be used when walnut pollen is hybridized;the male inflorescence was inactivated after 16 days of natural pollination,which provided a reference for its use as an pollination tree.
