摘 要:【目的】探明茶树害虫山香圆平背粉虱不同地理种群的遗传多样性,掌握其遗传分化现状,为其科学防治提供依据。【方法】基于贵州茶园山香圆平背粉虱10个地理种群的rDNA ITS1基因序列片段,采用MEGA-X、DnaSP 5.10、Arlequin 3.5.2.2及SAMOVA 2.0等研究其种群遗传分化、基因交流、分子变异及种群遗传多样性。【结果】贵州茶园山香圆平背粉虱10个地理种群的rDNA ITS1基因序列共获得68条基因序列片段,其序列长度为434 bp,包含保守位点306个,变异位点78个,简约信息位点48个,其中9个位点发生转换,9个位点发生颠换,具有明显的G+C偏倚性;其共定义47个单倍型,包括5个共享单倍型和42个独享单倍型;总群体遗传多样性较高,表现出高单倍型多样性(H_(d)=0.968)和高核苷酸多样性(P_(i)=0.03628);总群体遗传分化程度高(F_(st)=0.72460),基因交流水平低(N_(m)=0.10);遗传变异主要来自组间(F ct=0.64620);单倍型系统发育分析表明,山香圆平背粉虱聚为两分支,MT、YQ、FG与SY种群聚成一类,MT、DZ、RH、HX、QX、JS与HHG种群聚成一类。【结论】贵州茶园山香圆平背粉虱总体遗传多样性水平较高,基因交流水平较低,具有显著的遗传分化。【Objective】The genetic diversity of different geographical populations of the tea plant pest,Crenidorsum turpiniae,was explored to know the current status of genetic differentiation of C.turpiniae,and to provide the foundation for its scientific prevention and control.【Method】Based on the sequence fragments of rDNA ITS1 gene of 10 geographical populations of C.turpiniae in Guizhou Province,the genetic differentiation,gene flow,molecular variation and genetic diversity of these populations were analyzed using MEGA-X,DnasP 5.10,Arlequin 3.5.2.2,SAMOVA 2.0,etc.【Result】The sequence length of rDNA ITS1 gene of 10 geographical populations of C.turpiniae was 434 bp with obvious G+C bias,and it had 306 conservative sites,78 mutation sites and 48 parsimonious information site,among which 9 sites were converted and 9 site were reversed.A total of 47 haplotypes(5 haplotypes were shared and 42 haplotypes were exclusive)were defined.The genetic diversity of the total population of C.turpiniae showed high haplotype diversity(H_(d)=0.968)and high nucleotide diversity(P_(i)=0.03628)in Guizhou 10 plantations.The genetic differentiation degree of the total population of the 10 geographical populations of C.turpiniae was high(F_(st)=0.72460),as the gene flow was low(N_(m)=0.10).The genetic variation of the C.turpiniae was mainly from among groups(F ct=0.64620);The haplotype phylogenetic analysis showed that the C.turpiniae had two branches with one consisting of MT,YQ,FG and SY populations,and the other of MT,DZ,RH,HX,QX,JS and HHG populations.【Conclusion】The overall level of genetic diversity of C.turpiniae in Guizhou was high,gene glow was low,and it had significant genetic differentiation.
