秦皇岛地区克氏原螯虾源嗜水气单胞菌分离及生物学特征分析

摘  要:为鉴定引起克氏原螯虾死亡的病原菌及分析其生物学特征,从秦皇岛地区病死的疑似细菌感染的克氏原螯虾的肝脏和胰腺分离出1株病原菌,暂定名为ProcQHD。经细菌分离鉴定和16S rRNA基因序列分析,该菌株被鉴定为嗜水气单胞菌。通过人工感染动物试验、PCR法及K-B药敏纸片对其致病性、毒力基因、耐药基因及耐药性进行了研究。结果表明,ProcQHD株的LD_(50)为1.0×10^(5) CFU/mL,携带ser、act、exu、lip和Luxs 5种毒力基因;ProcQHD株对替米考星、头孢曲松、环丙沙星、土霉素、诺氟沙星等12种药物高度敏感且携带Tet(A)、Tet(C)、Tet(E)、qepA、gyrB、aac4、aadB、Ant-Ia、Sul-1、LAP-1、SHV 11种耐药基因。分析发现ProcQHD株的某些耐药表型与耐药基因之间有一定相关性。In order to identify the pathogenic bacteria causing the death of Procambarus clarkii and analyze its biological characteristics.A pathogen named ProcQHD was isolated from the liver and pancreas of dead P.clarkii.ProcQHD strain was identified as Aeromonas hydrophila by routine bacterial isolation,identification and 16S rRNA gene sequence analysis.The pathogenicity,virulence gene,drug resistance gene and drug resistance were detected by artificial infection animal experiment,PCR and K-B drug sensitivity test.The results showed that the LD_(50) of ProcQHD strain was 1.0×10^(5) CFU/mL,carried five virulence genes ser,act,exu,lip and LuxS;ProcQHD strain was highly sensitive to 12 kinds of drugs,such as tilmicosin,ceftriaxone,ciprofloxacin,oxytetracycline and norfloxacin.It carried 11 kinds of resistance genes,including Tet(A),Tet(C),Tet(E),qepA,gyrB,aac4,aadB,Ant-Ia,Sul-1,LAP-1,SHV,etc.There was some correlation between drug resistance phenotype and resistance gene.
