摘 要:目的:建立防瘟九味饮中(R,S)-告依春、葛根素、毛蕊异黄酮葡萄糖苷、升麻素苷、连翘酯苷A、5-O-甲基维斯阿米醇苷的LC-MS/MS定性分析方法及HPLC含量测定方法。方法:定性分析采用LC-MS/MS(配置ESI离子源)方法对6种活性成分进行确证;含量测定采用HPLC方法选取Agilent Zorbax SB C_(18)(150 mm×4.6 mm,5μm)色谱柱,流动相为0.2%磷酸水溶液和甲醇溶液,检测波长为254 nm,流速为1 mL/min,柱温30℃。结果:防瘟九味饮中6种活性成分在供试品溶液及对照品溶液中的质谱图相同,6种成分在各自浓度范围内线性关系良好,平均加样回收率99.54%~104.99%,RSD为1.24%~3.66%。结论:该方法操作简单快捷,灵敏度高,重复性好,研究结果可为防瘟九味饮的质量控制体系及医院制剂开发提供科学的参考依据。Objective To establish a LC-MS/MS method and a HPLC method for qualitative analysis and determination of(R,S)-Goitrin,puerarin,calycosin-7-O-β-D-glucosid,prim-O-glucosylcimifugin,forsythoside A,5-O-Methyl⁃visamminol in Fangwen Jiuwei Decoction.Methods Qualitative analysis was performed using LC-MS/MS(equipped with ESI ion source)method to confirm the six active components.Content determination was performed using HPLC method with Agilent Zorbax SB C18(150 mm×4.6 mm,5μm)column.The mobile phase was 0.2%phosphoric acid aqueous solu⁃tion and methanol solution.The detection was set as 254 nm,the flow rate was 1 mL/min and the column temperature was 30℃.Results The mass spectrograms of six active components in Fangwen Jiuwei Decoction were consistent with the refer⁃ence solution.The six components had good linear relationships in their respective concentration ranges.The average recov⁃eries were 99.54%~104.99%,and RSD values were 1.24%~3.66%.Conclusion The established method is simple and rapid with the good reproducibility and high sensitivity.The results can provide scientific reference for the quality control system of Fangwen Jiuwei Decoction and the development of hospital preparations.
