摘 要:目的:比较7种不同类型的癌组织与相应癌旁组织内胱抑素B(CSTB)的表达情况。方法:收集食管鳞状细胞癌、喉鳞状细胞癌、胃腺癌、肺鳞状细胞癌、肝细胞癌、结肠腺癌和膀胱移行细胞癌患者的癌组织与癌旁组织,体外培养食管癌Eca109细胞株、胃癌SGC7901细胞株、肝癌Hep G2细胞株及肺癌YTLMC-90细胞株,通过Western blot分析食管癌与相应癌旁粘膜组织内的CSTB表达水平,通过免疫组化法检测各组织标本和癌细胞株内CSTB的表达情况。结果:体外培养的癌细胞株中,食管癌Eca109细胞株、胃癌SGC7901细胞株、肝癌Hep G2细胞株及肺癌YTLMC-90均表达CSTB。在癌旁组织中,喉粘膜、食道粘膜、胃粘膜及肝组织表达CSTB,其它癌旁正常组织低表达或不表达CSTB;相应癌组织中,喉鳞状细胞癌、食管鳞状细胞癌及胃腺癌CSTB表达水平低(P<0.05);肝癌组织CSTB表达水平高(P<0.05),而肺鳞状细胞癌、结肠腺癌及膀胱移行细胞癌CSTB表达水平高(P<0.05)。结论:不同类型实体癌内CSTB的表达失调,为进一步阐释不同类型肿瘤发生发展机制提供分子基础。Objective:To compare the expression of cyatatin B(CSTS)in 7 different types of cancer tissues with corresponding adjacent tissues.Methods:Cancer tissues and paired adjacent non-cancerous tissues were collected from patients with esophageal squamous cell carcinoma,laryngeal squamous cell carcinoma,gastric adenocarcinoma,lung squamous cell carcinoma,hepatocellular carcinoma,colon adenocarcinoma,and bladder transitional cell carcinoma.The esophageal cancer cell line Eca109,gastric cancer cell line SGC7901,liver cancer cell line Hep G2,and lung cancer cell line YTLMC-90 were cultured in vitro.Western blot analysis was used to assess the expression levels of CSTB in esophageal cancer tissues and corresponding adjacent mucosal tissues.Immunohistochemistry was performed to detect the expression of CSTB in various tissue specimens and cancer cell lines.Results:CSTB was expressed in the cultured cancer cell lines,including Eca109,SGC7901,Hep G2,and YTLMC-90.Among the adjacent non-cancerous tissues,CSTB was expressed in adjacent laryngeal mucosa,esophageal mucosa,gastric mucosa,and liver tissues.Other adjacent non-cancerous tissues showed low expression or no expression of CSTB.However,in the corresponding cancer tissues,CSTB expression was decreased in laryngeal squamous cell carcinoma,esophageal squamous cell carcinoma,and gastric adenocarcinoma(P<0.05),while it remained highly expressed in liver cancer tissues(P<0.05).CSTB expression was increased in lung squamous cell carcinoma,colon adenocarcinoma,and bladder transitional cell carcinoma(P<0.05).Conclusion:These findings confirmed the dysregulation of CSTB expression in different types of solid cancers,providing a molecular basis for further elucidation of the mechanisms of different types of tumor development.
