摘 要:1-脱氧-D-木桐糖-5-磷酸合酶(DXS)是萜类化合物生物合成途径——甲基赤藓糖醇磷酸途径(MEP)中的一种必需酶。该酶是催化MEP途径的第一步反应,能够使丙酮酸和甘油醛3-磷酸缩合形成1-脱氧-D-酮糖-5-磷酸(DXP)。枯草芽孢杆菌遗传背景清晰,是一种优良的潜在萜类化合物生产菌。然而,关于枯草芽孢杆菌MEP途径关键酶DXS的研究仍不完善。本次研究克隆了枯草芽孢杆菌的dxs基因,构建了该基因的重组表达质粒,并将其转入枯草芽孢杆菌中进行过表达,分析DXS对枯草芽孢杆菌代谢途径的影响。结果表明,dxs基因的过表达能够引起枯草芽孢杆菌代谢途径异常,导致二氧化碳排放量增加。1-deoxy-D-xylulose-5-phosphate synthase(DXS)is an essential enzyme in the biosynthesis pathway of terpenoids,namely the 2-C-methyl-Derythritol 4-phosphate(MEP)pathway.This enzyme catalyzes the first step reaction of the MEP pathway,condensing pyruvate and glyceraldehyde-3-phosphate to form 1-deoxy-D-xylose 5-phosphate(DXP).Bacillus subtilis has a clear genetic background and is an excellent potential terpene producing bacterium.However,research on the key enzyme DXS in the MEP pathway of Bacillus subtilis is still insufficient.In this study the dxs gene of Bacillus subtilis was cloned,and a recombinant expression plasmid for this genewas also constructed.The recombinant expression plasmid was transferred into Bacillus subtilis for overexpression,and the effect of DXS on the MEP metabolic pathway in Bacillus subtilis was analyzed..The results showed that overexpression of the dxs gene caused abnormal metabolic pathways and increased carbon dioxide content in Bacillus subtilis.
