眼线百合花器官植株再生技术研究

摘　　要：以眼线百合的花器官为外植体进行组织培养,采用正交设计进行激素浓度配比,经过组织培养获得再生植株.结果表明:眼线百合花器官不同部位诱导情况不同,最容易诱导的为子房,其次为花丝,最难诱导的是花药.不同阶段的最优培养基有所不同,在诱导愈伤组织的过程中,花丝适宜培养基为MS+2,4-D 0.5 mg/L+NAA 0.1 mg/L+6-BA 1.0 mg/L,子房适宜培养基为MS+NAA 0.1 mg/L+6-BA 1.0 mg/L;在出芽诱导中,两种外植体最佳培养基均为MS+NAA 0.1 mg/L+6-BA 1.0 mg/L.在未添加激素的MS培养基中对试管苗进行壮苗培养,既可以诱导生根又可以达到壮苗的目的.眼线百合试管苗移栽基质配比为草炭∶沙∶蛭石∶珍珠岩=10∶2∶1∶1,成活率高达98%.The flower organs of Lilium‘Eyeliner’were used as explants for tissue culture,and the hormone concentration ratio was determined by orthogonal design.The regenerated plants were obtained through tissue culture.The results showed that the induction of different parts of the flower organs of Lilium‘Eyeliner’was different.The ovary was the most easily induced,followed by the filament,and the anther was the most difficult to induce.The optimal culture medium for different stages varies.During the process of inducing callus,the suitable culture medium for filaments is MS+2,4-D 0.5 mg/L+NAA 0.1 mg/L+6-BA 1.0 mg/L,and the suitable culture medium for ovaries is MS+NAA 0.1 mg/L+6-BA 1.0 mg/L.In bud induction,the optimal medium for both explants is MS+NAA 0.1 mg/L+6-BA 1.0 mg/L.Cultivating strong seedlings in MS medium without adding hormones can not only induce rooting but also achieve the goal of strong seedlings.The ratio of transplanting medium of test tube seedlings of Lilium‘Eyeliner’is peat:sand:vermiculite:Perlite=10∶2∶1∶1,and the survival rate is as high as 98%.
