摘  要:目的联合GEO数据库和网络药理学探讨鸡血藤-络石藤治疗强直性脊柱炎(ankylosing spondylitis,AS)的作用机制。方法采用TCMSP筛选出鸡血藤-络石藤的中药有效成分。检索DrugBank、GeneCards及DisGeNET数据库中关于AS的疾病靶点。从Gene Expression Omnibus(GEO)数据库筛选得到GSE25101的芯片数据集,并借助R软件4.2.2进行差异基因分析后,汇总至AS疾病基因靶点。利用Cytoscape 3.9.1软件构建“中药-有效成分-靶点”网络图,并通过String平台构建蛋白质相互作用网络,筛选得到核心靶点。利用DAVID数据库对交集基因进行富集分析。运用AutoDock Vina软件进行分子对接。结果本研究共筛选得到鸡血藤-络石藤治疗AS的靶点50个,有效成分31个,涉及IL17、TNF、炎症性肠病等多条信号通路。分子对接结果显示,木犀草素、刺芒柄花素等主要成分与CAT、MMP9、TNF、IL6等核心靶点有较好的结合潜力。结论鸡血藤-络石藤可能通过调控炎症反应等基因进而发挥治疗AS的作用。Objective To study the mechanism of spatholobi caulis-Chinese starjasmine stem in treating ankylosing spondylitis(AS)by combining GEO database and network pharmacology.Methods TCMSP was used to screen out the active ingredients of traditional Chinese medicine of spatholobi caulis-Chinese starjasmine stem.DrugBank,GeneCards,and DisGeNET databases were searched for disease targets for AS.The Gene Expression Omnibus(GEO)database screened the chip dataset of GSE25101 and summarized it to AS disease gene targets after differential gene analysis with the help of R software 4.2.2.Cytoscape 3.9.1 software was used to construct the network diagram of“traditional Chinese medicine-active ingredient-target”,and the protein interaction network was constructed through the String platform,and the core targets were screened.The DAVID database was used to enrich and analyze the intersecting genes.AutoDock Vina software was employed for molecular docking.Results In this study,a total of 50 targets and 31 active ingredients were screened for the treatment of AS by spatholobi caulis-Chinese starjasmine stem,involving multiple signaling pathways such as IL17,TNF,and inflammatory bowel disease.The results of molecular docking showed that the main components such as luteolin and stingular anthocyanin had good binding potential with core targets such as CAT,MMP9,TNF and IL6.Conclusion Spatholobi caulis-Chinese starjasmine stem may play a role in the treatment of AS by regulating genes such as inflammatory response.
