摘 要:目的研究增味小承气汤对梗阻性黄疸(OJ)大鼠肠道黏膜紧密连接蛋白-1(ZO-1)、分泌型免疫球蛋白A(s IgA)、黏蛋白-2(MUC-2)表达的影响,探讨增味小承气汤对肠黏膜屏障功能的保护作用机制。方法将SD雄性大鼠随机分为正常组、假手术组、手术建模梗阻性黄疸+中药组(OJ+M组)和手术建模梗阻性黄疸组(OJ组),每组10只。建立模型并进行实验干预,第3、5、7天自尾静脉取血。建立模型后第8天处死大鼠,留取标本,测量小肠绒毛高度及隐窝深度,免疫组化法检测ZO-1、s IgA、MUC-2的表达部位及表达量,观察增味小承气汤对肠黏膜机械屏障、免疫屏障、生物屏障功能的影响。结果与OJ组相比,OJ+M组的小肠绒毛高度增加,隐窝深度增加;OJ+M组ZO-1、s IgA、MUC-2的表达量更高。结论增味小承气汤增加了梗阻性黄疸大鼠肠黏膜ZO-1、s IgA和MUC-2的表达,减轻了肠粘膜屏障功能的损害。Objective To observe the effects of modified Xiaochengqi Decoction on the expression of the zonulaoccludens-1(ZO-1),secreted immunoglobulin A(s IgA)and mucoprotein-2(MUC-2)in the intestinal mucosa of rats with obstructive jaundice,and analyze the effects of modified Xiaochengqi Decoction on intestinal mucosal barrier function.Methods The SD male rats were randomly divided into the control group,the sham group,the surgical model of obstructive jaundice+modified Xiaochengqi Decoction(OJ+M)group and the surgical model of obstructive jaundice(OJ)group,with 10 rats in each group.Blood was collected via the caudal vein on the 3rd,5th and 7th day after modeling and experimental intervention.All rats were killed on the 8th day after modeling with corresponding specimens collected.The collected intestinal specimens were submitted for HE staining and their small intestinal villous height and crypt depth were measured;the expression sites and levels of ZO-1,s IgA and MUC-2 were detected by immunohistochemistry to reveal the effect of modified Xiaochengqi Decoction on intestinal mucosal mechanical barrier,immune barrier,and biological barrier functions.Results Compared with those in the OJ group,the small intestine villus height and the small intestine crypt depth were increased in the OJ+M group.The expression levels of ZO-1,s IgA,and MUC-2 were higher in the OJ+M group.Conclusion Modified Xiaochengqi Decoction increased the expression of ZO-1,s IgA and MUC-2 in intestinal mucosa of rats with obstructive jaundice,and alleviated the damage of intestinal barrier function.
