摘 要:为探讨车前草多糖(PLP)对高脂日粮(HF)饲喂引起的小鼠脂代谢异常和氧化应激的影响,本试验选取150只SPF级雄性健康昆明小鼠,适应性饲养1周后,随机分成5个组,分别为对照(CK)组、高脂日粮(HF)组、HF+100 mg/(kg·bw)PLP组、HF+200 mg/(kg·bw)PLP组和HF+400 mg/(kg·bw)PLP组。每天记录小鼠的采食量和饮水量,试验4周后收集其肝脏和血液进行脂代谢和氧化应激相关指标和基因测定。结果显示,与CK组相比,HF组小鼠血清中甘油三脂(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)和血糖(GLU)含量以及天冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)活性显著升高(P<0.05),高密度脂蛋白胆固醇(HDL-C)含量显著降低(P<0.05);肝脏中TC、TG、丙二醛(MDA)和活性氧自由基(ROS)含量显著升高(P<0.05),过氧化物酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活性均显著降低(P<0.05)。而添加200 mg/(kg·bw)和400 mg/(kg·bw)PLP显著降低了HF饮食小鼠血清中TG、TC、LDL-C和GLU含量以及AST和ALT活性(P<0.05),显著提高HDL-C含量(P<0.05);显著降低小鼠肝脏中TC、TG、MDA和ROS含量(P<0.05);显著提高肝脏中抗氧化酶CAT、GSH-Px和SOD活性(P<0.05)。此外,进一步对小鼠抗氧化和脂代谢相关基因检测发现,PLP可显著提高HF饮食导致的抗氧化基因Nrf 2和HO-1表达水平的降低,并降低脂代谢相关基因Lp l、Cyp7αl和Scd 1的表达(P<0.05)。结果表明,HF饮食可引发小鼠脂代谢异常和肝脏氧化应激并导致肝损伤,而PLP可缓解HF饮食导致的小鼠氧化应激和脂代谢异常,该结果可为PLP的临床应用提供参考依据。To investigate the impact of plantain polysaccharide(PLP)on lipid metabolism abnormalities and oxidative stress in mice fed with high-fat diet(HF),150 SPF male healthy Kunming mice were selected.After a week of adaptation,they were randomly divided into five groups:Control(CK)group,High-fat(HF)group,HF+100 mg/(kg·bw)PLP group,HF+200 mg/(kg·bw)PLP group,and HF+400 mg/(kg·bw)PLP group.Daily food and water intake were recorded,and after 4 weeks,liver and blood samples were collected for lipid metabolism and oxidative stress-related indicators and gene expression analysis.The results showed that compared with the CK group,mice in the HF group exhibited significantly elevated levels of serum triglycerides(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),and glucose(GLU),as well as increased aspartate transaminase(AST)and alanine transaminase(ALT)activities(P<0.05),and decreased high-density lipoprotein cholesterol(HDL-C)levels(P<0.05);liver TC,TG,malondialdehyde(MDA),and reactive oxygen species(ROS)levels were significantly increased(P<0.05),while the activities of catalase(CAT),glutathione peroxidase(GSH-Px),and superoxide dismutase(SOD)were significantly decreased(P<0.05).The addition of 200 mg/(kg·bw)and 400 mg/(kg·bw)PLP significantly decreased serum TG,TC,LDL-C,and GLU levels,as well as AST and ALT activities in HF fed mice(P<0.05),and significantly increased HDL-C levels(P<0.05),significantly reduced liver TC,TG,MDA,and ROS levels(P<0.05),while enhancing the activities of antioxidant enzymes CAT,GSH-Px,and SOD(P<0.05).Furthermore,gene expression analysis related to antioxidant and lipid metabolism in mice revealed that PLP significantly upregulated the reduced expression levels of antioxidant genes Nrf 2 and HO-1 induced by the HF diet and decreased the expression of lipid metabolism-related genes Lp l,Cyp 7α1,and Scd 1(P<0.05).These results suggest that the HF diet can induce lipid metabolism abnormalities and hepatic oxidative stress,leading to liver damage in mice.However,PLP can all
