摘  要:研究了中药赤芍中的单萜类化合物paeonilactone B的体外抗炎活性及其作用机制.通过脂多糖(LPS)诱导RAW264.7单核巨噬细胞建立体外抗炎模型,发现paeonilactone B能显著抑制LPS诱导的巨噬细胞中NO的产生,IC 50值为6.09±1.81μmol/L.对paeonilactone B进行抗炎机制研究,发现paeonilactone B能抑制LPS激活的巨噬细胞中细胞因子IL-1β、IL-6、IL-18和TNF-α的分泌和炎症相关蛋白酶iNOS和COX-2的生成,同时,能抑制信号通路蛋白(JNK、ERK1/2、p38MAPK、IκBα、NF-κB/p65)的磷酸化;另外,paeonilactone B对流感病毒A/WSN/33/2009 H1N1(WSN)感染的巨噬细胞中的NO、TNF-α和IL-18产生也具有抑制作用,同时降低iNOS和COX-2的表达,但对IL-1β、IL-6的分泌没有影响.实验结果表明:paeonilactone B在LPS激活的巨噬细胞中可以通过抑制MAPKs和NF-κB信号通路来降低炎症因子的分泌而发挥抗炎活性;此外,对流感病毒感染的巨噬细胞也表现出一定的抗炎潜力.This study investigates the anti-inflammatory activity and mechanism of paeonilactone B found in the traditional Chinese medicine Paeoniae Radix Rubra in vitro.An in vitro anti-inflammatory model of RAW264.7 macrophages induced by lipopolysaccharide(LPS)was established.It was found that paeonilactone B significantly inhibited the production ofnitric oxide(NO)in LPS induced macrophages with IC 50 value of 6.09±1.81μmol/L.The anti-inflammatory mechanism research showed that paeonilactone B inhibited the secretion of cytokines IL-1β,IL-6,IL-18 and TNF-αand reduced the expression of iNOS and COX-2 on LPS-induced macrophages.Additionally,it suppressed the phosphorylation of signaling pathway proteins(JNK,ERK1/2,p38MAPK,IκBα,NF-κB/p65).Furthermore,paeonilactone B also decreased the level of NO,TNF-α,IL-18 and the expression of iNOS and COX-2 in macrophages infected with influenza virus A/WSN/33/2009 H1N1(WSN),but had no effect on IL-1βand IL-6.The results indicate that paeonilactone B exerts its anti-inflammatory activity in LPS-activated macrophages by inhibiting MAPKs and NF-κB signaling pathways to reduce the secretion of inflammatory factors.Additionally,it demonstrates potential anti-inflammatory effects in macrophages infected with the influenza virus.
