摘  要:根际微生物对水库消落带植被的生长繁殖、营养循环及适应性具有直接影响.然而,白鹤滩水库消落带由于地处偏远、形成时间不长,因而对其优势植物根际微生物的组成、多样性及代谢功能尚不明确.本研究以正常(YA)和萎蔫(YB)银胶菊(Parthenium hysterophorus L.)植株根际土壤为对象,采用Illumina Novaseq高通量测序分析其微生物多样性.结果表明,YA和YB的古细菌丰度一致,且都由Candidatus Nitrososphaera构成.YA和YB的最优势细菌菌属同为Pseudomonas,但是次优的细菌菌属的种类则不相同,它们共有的细菌ASV达到1246个,特有的分别为2130个和2777个.YA和YB的最优势真菌菌属并不相同,YA中Wickerhamomyces是最丰富的菌属,而YB中最优势真菌菌属则为Coprinellus,共有的真菌ASV为209个,独有的分别为564个和308个.通过LEfSe分析,本研究发现了YA和YB中差异最显著的关键分类单元.微生物代谢功能分析显示原核微生物和真菌相对丰度最高的代谢通路分别与有氧呼吸和棕榈酸/腺苷核糖核苷酸从头生物合成有关.该研究对理解长江上游大型水库消落带优势植被与微生物互作具有理论和指导意义.Rhizosphere microorganisms have a direct impact on the growth,reproduction,nutrient cycle and adaptability of the vegetation in the water-level-fluctuation zone of the reservoirs.However,the composition,diversity and metabolic functions of rhizosphere microorganisms of dominant plants in the water-level-fluctuation zone of Baihetan Reservoir are still unclear due to its remote location and short formation time.In this study,Illumina Novaseq high-throughput sequencing was used to analyze the microbial community diversity of rhizosphere soil of normal(YA)and wilted(YB)Parthenium hysterophorus L.The results show that the abundance of archaea in YA and YB is consistent,and both of them are composed of Candidatus Nitrososphaera.The most dominant bacterial genus of YA and YB is Pseudomonas,but other bacterial genera have changed largely,with 1246 bacteria ASV being common in them,and 2130 and 2777 being unique in YA and YB,respectively.The most dominant fungal genera of YA and YB are different.In YA,Wickerhamomyces is the most abundant fungal genus,while in YB,Coprinellus is the most dominant fungal genus,with 209 fungi ASV being common in them,and 564 and 308 being unique in YA and YB,respectively.The key taxon that contributes the most to the observed differences in YA and YB was identified by LEfSe analyses.It was shown that the metabolic pathway with the highest abundance of prokaryotes and fungi was PWY-3781 and PWY-5994/PWY-7219,respectively.This study has theoretical and guiding significance for understanding the interaction between dominant vegetation and microorganisms in the water-level-fluctuation zone of large reservoirs in the upper reaches of the Yangtze River.
