摘  要:目的:探讨不同浸提介质(去离子水、生理盐水、其他介质)对一次性使用血液灌流器体外细胞毒性评价的影响。方法:在研究时间段内(2023年2月至5月),用MTT法测定了不同血液灌流器的细胞毒性。将血液灌流器浸泡在三种介质中,按国家药典委员会要求提取。用提取液替换培养基,与L929细胞共培养4h。加入MTT溶液,测定紫色产物的吸光度值,计算相对存活率,评估细胞毒性。结果:不同浸提介质对血液灌流器细胞毒性的影响不同。其中,0.9%氯化钠浸提时的相对存活率最高,培养基次之,含血清培养基浸提时的相对存活率最低。这可能是因为0.9%氯化钠浸提时,血液灌流器释放出的物质较少,对细胞的损伤较小;而含血清培养基浸提时,血液灌流器释放出的物质较多,对细胞的损伤较大。结论:不同浸提介质对一次性使用血液灌流器体外细胞毒性评价有显著影响,应根据实际应用场景选择合适的浸提介质进行评价。Objective:To investigate the effects of different extraction media(deionized water,normal saline,other media)on the in vitro cytotoxicity evaluation of disposable hemoperfusion device.Methods:During the study period(February to May 2023),the cytotoxicity of different hemoperfusion devices was measured by MTT assay.The hemoperfusion device was immersed in three media and extracted as required by the National Pharmacopoeia Commission.The medium was replaced with extracts and co-cultured with L929 cells for 4 hours.MTT solution was added,the absorbance values of the purple product were determined,relative viability was calculated and cytotoxicity was assessed.Results:Different extraction media had different effects on the cytotoxicity of the hemoperfusion apparatus.The highest relative survival was achieved with 0.9%sodium chloride,followed by culture medium and lowest in serum-containing medium.This may be because the 0.9%sodium chloride releases less material and causes less damage to the cells;With serum medium,the device releases more material and more damage to the cells.Conclusion:Different extraction media have a significant influence on the in vitro cytotoxicity evaluation of disposable hemoperfusion,so appropriate extraction media should be selected according to the actual application scenario.
