摘 要:目的:研究硒代蛋氨酸(selenomethionine,SeMet)对胶原诱导型关节炎(collagen-induced arthritis,CIA)大鼠成纤维样滑膜细胞(fibroblast-like synoviocytes,FLS)中炎症因子释放和细胞内谷胱甘肽过氧化物酶(GSH-Px)活性的影响。方法:复苏CIA-FLS MH7A细胞系,传代培养后加入不同浓度的L-硒代蛋氨酸(0.1µmol/L,1µmol/L,10µmol/L,100µmol/L,500µmol/L,1000µmol/L)干预。通过MTT比色法,观察不同浓度L-硒代蛋氨酸对CIA-FLS细胞增殖的影响,后续采用比色法检测L-硒代蛋氨酸对CIA-FLS细胞内GSH-Px活性的影响;采用酶联免疫吸附法(Enzyme linked immunosorbent assay,Elisa)检测干预后的CIA-FLS细胞上清液中肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)的含量。结果:与空白对照组比较,0.1µmol/L L-硒代蛋氨酸溶液在24 h和48 h内对CIA-FLS细胞的增殖活性无明显影响。与空白对照组比较,0.1µmol/L的L-硒代蛋氨酸溶液作用24 h和48 h后,可有效提高CIA-FLS细胞内GSH-Px活性(P<0.05),且给药48 h后对FLS细胞内GSH-PX活性的保护作用更显著;0.1µmol/L的L-硒代蛋氨酸干预48 h后,可抑制CIA-FLS细胞中炎症因子TNF-α和IL-6的释放(P<0.05)。结论:L-硒代蛋氨酸能提高体外培养的CIA-FLS细胞内GSH-Px的活性并抑制其释放炎症因子,为后期进一步研究补硒治疗类风湿关节炎的作用机制提供实验依据。Objective:To investigate the effects of selenomethionine(SeMet)on the release of inflammatory factors and the activity of intracellular glutathione peroxidase(GSH-Px)in fibroblast-like synoviocytes(FLS)of rats with collagen-induced arthritis(CIA).Methods:The CIA-FLS MH7A cell line was resuscitated,and after subculture,L-SeMet at different concentrations(0.1µmol/L,1µmol/L,10µmol/L,100µmol/L,500µmol/L,1000µmol/L)was added for intervention.The effects of L-SeMet at different concentrations on the proliferation of CIA-FLS cells were observed by MTT colorimetry,and then the effects of L-SeMet on the GSH-Px activity in CIA-FLS cells were detected by colorimetry.The contents of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in the supernatant of CIA-FLS cells after intervention were detected by Enzyme linked immunosorbent assay(Elisa).Results:Compared with the blank control group,0.1µmol/L L-SeMet solution had no significant effect on the proliferation of CIA-FLS cells within 24 h and 48 h.Compared with the blank control group,0.1µmol/L L-SeMet solution could effectively increase the intracellular GSH-Px activity of CIA-FLS cells after treatment for 24 h and 48 h(P<0.05),and the protective effect on the activity of GSH-PX in FLS cells was more significant after 48 h of administration.After 48 h intervention with 0.1µmol/L L-SeMet,the release of inflammatory factors TNF-αand IL-6 in CIA-FLS cells was inhibited(P<0.05).Conclusion:L-SeMet can increase the activity of GSH-Px and inhibit the release of inflammatory factors in CIA-FLS cells cultured in vitro,which provides an experimental basis for further study of the mechanism of selenium supplement in the treatment of rheumatoid arthritis.
