摘  要:血管内皮细胞因解剖部位不同,在结构和功能等方面存在明显异质性。最新研究表明真皮微血管内皮细胞在代谢、信号转导等方面具有独特的表现。不同组织来源的血管内皮细胞之间是不可相互替代的,因此利用原代真皮微血管内皮细胞开展皮肤疾病研究更具有意义。但目前真皮微血管内皮细胞的获取仍有难度,存在不易分离、纯度较低、易发生成纤维细胞污染等困难。本文归纳了目前已发表的在正常与多种病理状态下皮肤微血管内皮细胞的分离、纯化以及鉴定方法并分析了优缺点,旨在为读者选择合适的皮肤微血管内皮细胞原代培养方法提供参考。Vascular endothelial cells have significant heterogeneity in structure and function due to different anatomical sites.Recent studies have shown that dermal microvascular endothelial cells have unique manifestations in metabolism and signal transduction.Vascular endothelial cells derived from different tissues cannot replace each other,so primary dermal microvascular endothelial cells are more meaningful for the study of skin diseases.However,at present,it is still difficult to obtain dermal microvascular endothelial cells,and there are difficulties such as difficult separation,low purity,and high proneness to generate fibroblast pollution.This paper summarized the methods of isolation,purification and identification of skin microvascular endothelial cells under normal and various pathological conditions,and analyzed their advantages and disadvantages,in order to provide a reference for readers to choose a suitable method for primary culture of skin microvascular endothelial cells.
