摘 要:宁粳50号是宁夏黄河灌区的主栽品种,生产上表现产量高、稻瘟病抗性好,稻米品质优,但缺乏香味。利用基因编辑技术对宁粳50号的香味基因BADH2进行定向敲除,获得了3个阳性克隆。经过田间综合农艺性状的对比和筛选,经4代自交选育出生育期、有效穗数、株高等综合农艺性状与宁粳50号相当,稻米具有明显香味,精米垩白粒率和垩白度等品质性状无明显改变的改良株系T1、T2和T3,实现了对宁粳50号的稻米香味性状的定向改良。Ningjing 50 is the main variety planted in the Yellow River irrigation area of Ningxia.Ningjing 50 exhibits high yield,good resistance to rice blast,and excellent rice quality,but lacks fragrance of rice.Three positive clones were obtained by targeted knockout of the fragrance gene BADH2 in Ningjing 50 using CRISPR/Cas9 gene editing technology.After comparing and screening the comprehensive agronomic traits in the field,after 4 generations of selfing,the improved lines T1,T2,and T3 with significant fragrance and no significant changes in rice chalkiness rate and chalkiness degree were selected,and the comprehensive agronomic traits of growth period,effective panicle number,and plant height were comparable to Ningjing 50.This study achieved targeted improvement of the fragrance traits of Ningjing 50.
