网络药理学研究升白方治疗免疫力低下的作用机制

摘  要:目的基于网络药理学探究升白方对环磷酰胺诱导小鼠免疫抑制模型的治疗机制。方法建立环磷酰胺免疫抑制模型,观察升白方对小鼠免疫损伤的影响。采用网络药理学分析建立“疾病-药物-靶点”网络,确定生物学过程和信号通路;采用Western blot方法对代表性通路进行实验验证。结果升白方显著逆转了免疫抑制小鼠体重、胸腺指数、脏器指数、碳粒廓清指数、吞噬指数、足跖增厚、巨噬细胞吞噬功能及自然杀伤细胞杀伤活性的变化。网络药理学预测出升白方含有134个生物活性成分、577个作用靶点、133个与免疫力低下的共同靶点;得到了6个主要活性因子,分别为TP53、AKT1、JUN、MAPK1、MAPK3、RELA。Western blot分析表明:升白方显著促进NF-κB的表达。结论升白方可显著增强免疫抑制小鼠的免疫功能,其可能通过激活NF-κB信号通路发挥增强机体免疫力活性的作用,有望用于化疗后免疫抑制保健产品的开发。OBJECTIVE To explore the therapeutic mechanism of Shengbai formula on cyclophosphamide induced immunosuppression in mice based on network pharmacology.METHODS A cyclophosphamide immunosuppressive model was established to observe the effect of Shengbai formula on immune damage in mice.A"disease drug target"network was establishesd by Network pharmacology analysis to identify biological processes and signaling pathways,and Western blot methods was used to experimentally validate representative pathways.RESULTS Shengbai formula significantly reversed the changes of body weight,thymus index,organ index,carbon particle clearance index,phagocytosis index,foot plantar thickening,macrophage phagocytosis and natural killer cell killing activity in immunosuppressed mice.Network pharmacology predicted that Shengbai formula contained 134 bioactive components,577 action targets,and 133 common targets with immune deficiency.Six main active factors were identified as TP53,AKT1,JUN,MAPK1,MAPK3,and RELA,respectively.Western blot analysis showed that Shengbai formula significantly promoted NF-κB expression.CONCLUSION Shengbai formula can significantly enhance the immune function of immunosuppressed mice,possibly by activating NF-κB.The signaling pathway plays a role in enhancing the body's immune activity and is expected to be used for the development of immunosuppressive health products after chemotherapy.
