罗汉果茎段组织培养与植株再生体系的研究

摘　　要：以青皮罗汉果茎段为外植体,探讨了不同消毒剂、消毒时间、不同丝瓜络提取物浓度和不同激素组合对罗汉果茎段组培育苗的影响,以期能够在较短时间内培育出大量优质种苗,加快推广速度,同时也是解决罗汉果病毒病的根本途径。结果表明:以罗汉果茎段作为外植体,其最佳消毒方式是0.05%的高锰酸钾溶液浸泡消毒25 min,0.1%的升汞消毒8 min,75%的乙醇消毒30 s,成活率达81%。芽诱导最佳培养基为MS+2.0 mg/L丝瓜络提取物浓度,诱导率达92%;芽增值最佳培养基为MS+1.0 mg/L丝瓜络提取物浓度+0.1 mg/L NAA,增殖系数达2.43;生根培养的最佳培养基为1/2 MS+0.1 mg/L NAA+0.1 mg/L IBA+0.2 mg/L碳粉,生根数在5~9条之间,生根率高达93%;最佳炼苗基质组成为:蛭石∶珍珠岩∶苔藓体积比=2∶1∶1,移栽成活率达100%。The effect of different disinfectants,disinfection time,concentration of luffa extract and hormone combinations onstem tissue culture of Siraitia grosvenorii were studied,so as to cultivate a large number of high-quality seedlings in a short time.The results showed that the best disinfection method was to use 0.05%potassium permanganate solution for 25 min,or 0.1%mercury solution for 8 min,or 75%ethanol for 30 s,under which conditions the survival rate reached 81%.The best medium for bud induction was MS plus 2.0 mg/L luffa extract,and the induction rate was 92%.The best medium for bud multiplication was MS plus 1.0 mg/L luffa extract plus 0.1 mg/L NAA,and the multiplication coefficient was 2.43.The best medium for rooting culture was 1/2 MS plus 0.1 mg/L NAA plus 0.1 mg/L IBA plus 0.2 mg/L carbon powder.The rooting number was between 5 and 9,and the rooting rate was as high as 93%.The optimum transplant substrate composition was as follows:the volume ratio of vermiculite,perlite and bryophyte was 2∶1∶1,and the survival rate reached 100%.
