摘  要:目的:探究坍塌反应调节蛋白2对七氟醚诱导的发育大鼠远期记忆功能障碍的影响。方法:构建CRMP2靶基因的野生型腺病毒,进行SD幼鼠海马定位注射,应用Western Blot、免疫荧光验证过表达效果。腺病毒注射后将幼鼠养至第七天,进行空气与七氟醚暴露4 h,31-32天时进行场景恐惧记忆实验。结果:在场景恐惧记忆实验中,七氟醚暴露后的大鼠的“木僵”时间明显降低。然而,过表达CRMP2能够逆转七氟醚所致的大鼠“木僵”时间的下降,提示七氟醚能够导致大鼠远期记忆功能损害,而CRMP2能够逆转七氟醚所致的记忆功能障碍。结论:七氟醚诱导新生大鼠远期记忆功能障碍的机制可能与抑制CRMP2表达有关。Objective:To explore the effect of collapse response regulatory protein 2 on long-term memory dys-function induced by sevoflurane in developing rats.Methods:Wild type adenovirus with CRMP2 target gene was con-structed and injected into the hippocampus of young SD mice.The overexpression effect was verified by Western Blot and immunofluorescence.After adenovirus injection,the young mice were raised until the seventh day,and were exposed to air or sevoflurane for 4 hours.The scene fear memory experiment was performed at 31-32 days.Results:In the scene fear memory experiment,the“freezing”time of rats was significantly reduced after sevoflurane exposure.However,Overexpression of CRMP2 can reverse the decrease of“freezing”time induced by sevoflurane in rats,suggesting that sevoflurane can cause long-term memory impairment in rats,while CRMP2 can reverse the sevoflurane-induced memory dysfunction.Conclusion:The inhibition of CRMP2 expression may be one of the mechanisms of sevoflurane-induced long-term memory impairment in neonatal rats.
