摘　　要：目的:采用两样本孟德尔随机化研究方法评估偏头痛与子宫内膜异位症之间的潜在因果关系。方法:研究数据来自全基因组关联研究(GWAS)汇总数据集,其中偏头痛数据包含8547例患者和176107例对照,子宫内膜异位症8288患者和68969例对照,选取7个与偏头痛密切相关的遗传位点作为工具变量。分别使用随机效应逆方差加权(IVW)、MR-Egger和加权中位数等方法进行两样本孟德尔随机化分析,采用优势比(OR)评估偏头痛与子宫内膜异位症之间的因果关系。最后辅以各种敏感性分析实验检验评估结果的有效性和鲁棒性。结果:IVW等4种分析方法均显示偏头痛与子宫内膜异位症呈正相关。其中IVW方法的OR为1.225(95%CI 1.133~1.325;P=0.009),其他3种方法也获得了相似的因果关系。此外,Cochrane s Q检验、MR-Egger回归截距(P=0.860)和漏斗图显示孟德尔随机化实验没有异质性和水平多效性。结论:偏头痛与子宫内膜异位症之间可能存在正向因果关系。Objective:To evaluate the potential causal relationship between migraine and endometriosis.Methods:The data were collected from a pooled Genome-wide Association Study(GWAS)dataset with 8547 patients and 176107 controls for migraine and 8288 patients and 68969 controls for endometriosis.There were 7 genetic loci strongly associated with migraine selected as instrumental variables.Two-sample Mendelian randomization analysis was performed using random-effect inverse-variance-weighting(IVW),MR-Egger and weighted median methods,respectively.Odds ratio(OR)was used to evaluate the causal relationship between migraine and endometriosis.Finally,a variety of sensitivity analysis experiments were performed to verify the effectiveness and robustness of the estimation results.Results:The results showed a positive association between migraine and endometriosis.The IVW method had an OR of 1.225(95%CI 1.133-1.325;P=0.009),and similar causal relationships were also obtained by other three methods.In addition,the MR-Egger regression intercept(P=0.860)and funnel plot showed no heterogeneity and horizontal pleiotropy in the Mendelian randomization experiment.Conclusion:There may be a positive causal relationship between migraine and endometriosis.
