摘 要:目的确定从真菌CHS3发酵产物中提取柴胡皂苷d的最优工艺。方法以柴胡皂苷d产量为指标,利用单因素分析与响应面分析相结合,优化乙醇浓度、料液比、超声功率、超声时间和超声温度的实验条件。结果真菌CHS3发酵产物中提取柴胡皂苷d的最佳提取条件为:72%的乙醇超声提取,料液比为1∶22(g∶m L),超声功率为300W,超声温度为56℃,超声时间为28min。该工艺条件下柴胡皂苷d的平均产量为3.928μg·g^(-1)。结论所得工艺条件稳定可靠,研究结果为利用发酵工程生产SSd提供理论与实验参考。OBJECTIVE The optimal process for extracting saikosaponin d from the CHS3 strain fermentation products was determined.METHOD The experimental conditions of ethanol concentration,material-liquid ratio,ultrasonic power,ultrasonic time and ultrasonic temperature were optimized by single factor analysis and response surface analysis,based on the yield of saikosaponin d.RESULTS The optimal extraction conditions of saikosaponin d from CHS3 strain fermentation products were as follows:ultrasonic extraction with 72%ethanol,solid-liquid ratio of 1:22,300W,28min,56℃.The average SSd yield under this process condition was 3.928μg·g^(-1).CONCLUSION The optimized process conditions are stable and reliable.The results provide theoretical and experimental reference for the production of SSd by fermentation engineering.
