补肾温阳化瘀方对子宫内膜异位症大鼠PKC信号通路的影响

摘　　要：目的:研究补肾温阳化瘀方治疗子宫内膜异位症(EMs)的作用,探讨其调控蛋白激酶C(PKC)信号通路的机制。方法:将50只SPF级8~10周龄雌性SD大鼠随机分为5组,分别为假手术组、模型组、中药低剂量组、中药高剂量组和西药组,每组10只。采用自体移植法复制EMs大鼠模型,造模成功后分别给予补肾温阳化瘀方低、高剂量及孕三烯酮灌胃,假手术和模型组灌胃蒸馏水,连续21 d。HE染色观察在位内膜及异位灶的病理形态改变;Western blot测定在位内膜及异位灶磷酸化蛋白激酶C的α亚型(p-PKCα)、蛋白激酶C的α亚型(PKCα)蛋白的表达水平。结果:模型组在位内膜和异位灶p-PKCα蛋白水平升高;给予补肾温阳化瘀方治疗后,p-PKCα的蛋白表达水平降低(P<0.05),PKCα的蛋白表达水平无明显变化。结论:补肾温阳化瘀方通过抑制PKC信号通路来治疗EMs。Objective:To study the effect of Bushen Wenyang Huayu(kidney-tonifying,yang-warming and stasis-removing)Formulaformula in the treatment of endometriosis(EMs),and explore its mechanism of regulating the protein kinase C(PKC)signaling pathway.Methods:508-10-week-old female SD rats of SPF grade were randomly divided into 5 groups:sham surgery group,model group,low-dose Chinese medicine group,high-dose Chinese medicine group,and western medicine group,with 10 rats in each group.Autologous transplantation method was used to replicate the EMs rat model.After successful modeling,low and high doses of Bushen Wenyang Huayu Formula,as well as progesterone,were orally administered while the sham surgery and model groups were orally administered distilled water,for 21 consecutive days.HE staining was used to observe the pathological morphological changes of eutopic endometrium and ectopic lesions;Western blot to determine the expressions of phosphorylated protein kinase CαSubtype in eutopic endometrium and ectopic lesions(p-PKCα)and protein kinase CαSubtype(PKCα).Results:p-PKCαof eutopic endometrial and ectopic lesions in the model group elevated;After treatment with Bushen Wenyang Huayu Formula,p-PKCαexpression decreased(P<0.05),and there was no significant change in the protein expression of PKCα.Conclusion:Bushen Wenyang Huayu Formula treats EMs by inhibiting the PKC signaling pathway.
