摘  要:目的优化黄毛耳草总黄酮的提取工艺,测定黄毛耳草药材中黄酮类成分芦丁、烟花苷的含量。方法考察乙醇浓度、提取时间、液料比3个因素,采用Box-Behnken响应面法设计黄毛耳草总黄酮提取条件并优化提取工艺。采用HPLC法测定黄毛耳草药材中芦丁、烟花苷的含量,以甲醇-0.5%甲酸溶液为流动相,检测波长为360 nm,流速1.0 mL/min,柱温为30℃。结果黄毛耳草总黄酮的最佳提取工艺为:乙醇浓度70%、超声时间47 min、液料比12.5∶1,此条件下黄毛耳草总黄酮得率为12.62%,与预测值12.59%差异无统计学意义。黄毛耳草中芦丁的含量为0.74 mg/g,烟花苷的含量为0.058 mg/g。结论利用Box-Behnken响应面设计法优化的黄毛耳草总黄酮提取工艺方法稳定,预测性较好,可为黄毛耳草总黄酮的提取工艺提供参考。建立的高效液相色谱法操作简便、结果准确,可用于同时测定黄毛耳草中芦丁和烟花苷的含量。Objective To optimize the extraction process of total flavonoids from hedyotis chrysotricha and determine the content of flavonoid components such as rutin and kaempferol-3-O-rutinoside in hedyotis chrysotricha.Methods The method takes ethanol concentration,extraction time,and liquid-solid ratio as three factors to investigate.The Box Behnken method was used to design the extraction conditions and optimize the extraction process of total flavonoids from hedyotis chrysotricha.Using methanol 0.5%formic acid solution as the mobile phase;360 nm was the detection wavelength;1.0 mL/min was the flow rate;The column temperature was 30℃,and the content of rutin and kaempferol-3-O-rutinoside in hedyotis chrysotricha was determined.Results The optimal extraction process for total flavonoids from hedyotis chrysotricha was as follows:ethanol concentration of 70%,ultrasound time of 47 minutes,and solid-liquid ratio of 12.5∶1.Under these conditions,the yield of total flavonoids from herba hedyotids chrysotrichae was 12.62%,which was not significantly different from the predicted value of 12.59%.The content of rutin in hedyotis chrysotricha was 0.74 mg/g,and the content of kaempferol-3-O-rutinoside was 0.058 mg/g.Conclusion The optimization of the extraction process of total flavonoids from hedyotis chrysotricha using Box Behnken response surface design method is stable and has good predictability,which can provide reference for the extraction process of total flavonoids from hedyotis chrysotricha.The established HPLC method is simple to operate and accurate in results,and can be used for the simultaneous determination of rutin and kaempferol-3-O-rutinoside in hedyotis chrysotricha.
