摘  要:目的研究大鼠同种异体骨基质明胶(bone matrix gelatin,BMG)吸附骨髓间充质干细胞(mesenchymal stem cell,MSCs)修复骨缺损的可行性及有效性。方法每组18只大鼠,对照组BMG植入,实验组MSCs经荧光标记后与BMG共同培养植入。均不予以内外固定,逐层缝合伤口。分别于术后1、2、3周、1、2个月处死大鼠2只。比较各组成骨效应。结果术后2个月取骨缺损区组织进行组织学观察,实验组采用荧光染料标记确认骨缺损区的骨痂是否来源于MSCs。对照组类骨样组织形成量较实验组同期标本明显较少,且骨量与成熟程度均明显低于实验组。荧光染料标记确认胫骨骨缺损区的骨痂来源于MSCs。与对照组比较,实验组1、2、3周、1、2个月放射学评分较高,差异有统计学意义(P<0.05)。与对照组比较,实验组1、2、3周、1、2个月组织学评分较高,差异有统计学意义(P<0.05)。结论大鼠BMG吸附MSCs修复骨缺损具有可行性及有效性。Objective To study the feasibility and effectiveness of bone marrow mesenchymal stem cells(MSCs)absorbed by allogeneic bone matrix gelatin(BMG)in repairing bone defects.Methods eighteen rats in each group were divided into control group(BMG implantation)and experimental group(MSCs were co cultured and implanted with BMG after fluorescent labeling).The wounds were sutured layer by layer without internal and external fixation.Two rats were killed at 1 week,2 weeks,3 weeks,1 month and 2 months after operation respectively to compare the bone effect of each component.Results 2 months after operation,the tissue of bone defect area was taken for histological observation.In the experimental group,fluorescent dye labeling was used to confirm whether the callus in the bone defect area came from MSCs.The amount of osteoid tissue formation in the control group was significantly less than that in the experimental group,and the bone mass and maturity were significantly lower than those in the experimental group.Fluorescent dye labeling confirmed that the callus in the tibial bone defect area came from MSCs.Compared with the control group,the experimental group had higher radiological scores at 1 week,2 weeks,3 weeks,1 month,and 2 months,and the difference was statistically significant(P<0.05).Compared with the control group,the experimental group had higher histological scores at 1 week,2 weeks,3 weeks,1 month,and 2 months,and the difference was statistically significant(P<0.05).Conclusion MSCs adsorbed by BMG is feasible and effective in repairing bone defects.
