摘 要:甲胎蛋白(AFP)被认定为肝癌的生物标志物,因此开发出一种操作简单、灵敏度高的AFP检测手段对肝癌的检测具有重要的临床意义。本研究设计了无标记电位型溶液栅石墨烯晶体管(SGGT)生物传感器,在栅电极上电沉积纳米金用来捕获抗体AFP,使用牛血清白蛋白(BSA)来封闭活性位点,最终抗原AFP被抗体特异性识别结合在电极上。在该研究中,抗体AFP、BSA和AFP的等电点均小于中性的PBS测试液,因此带负电荷,导致界面电势发生变化,最终引起转移曲线中性点的偏移。实验表明该生物传感器对甲胎蛋白的检测具有较好的线性关系:ΔI_(DS)=45.42lg c+4.98,在5~200 ng/mL的检测范围内,检测限低至1.00 ng/mL。该方法为甲胎蛋白的检测分析提供了一个可行的方法,并期望构建便携式传感器,实现高通量的分析检测。Alpha fetoprotein(AFP)is widely accepted as the biomarker of liver cancer,so it is of great clinical significance to develop detection of liver cancer methods with simple operation and high sensitivity.A label-free potentiometric solution gate graphene transistor(SGGT)biosensor was designed,and the gold nanoparticles were electrodeposited on the gate electrode to capture antibody AFP.The bovine serum albumin(BSA)blocked the other active sites.Here,the AFP was specifically recognized and bounded to the electrode by the antibody.In the transistor system,the isoelectric points of the antibody AFP,BSA and AFP were all smaller than that of the neutral PBS test solution,so they were negatively charged,resulting some changes in the interface potential,and eventually causing the shift of the neutral point of the transfer curve.The experiments showed that the biosensor had a good linear relationship for the detection of alpha-fetoprotein in the detection range of 5 ng/mL~200 ng/mL(the detection limit was 1.00 ng/mL),ΔI _(DS)=45.42lg c+4.98.This work provides a feasible way for the detection of the alpha-fetoprotein,which is expected to construct a portable sensor to achieve high-throughput detection.
