摘 要:目的探讨高糖高脂对人主动脉内皮细胞(HAECs)的影响及通心络的干预作用。方法经随机方式,将HAECs分为以下5组:通心络高、中、低浓度组,高糖高脂组和空白对照组。检测各组细胞生存活性、细胞上清液乳酸脱氢酶(LDH)含量、三磷酸腺苷(ATP)含量、线粒体膜电位(MMP)、细胞内活性氧簇(ROS)水平和细胞色素(Cyt)-C蛋白表达。结果与空白对照组比较,高糖高脂组MMP、ATP水平与细胞生存活性明显下降,细胞中ROS含量、细胞上清液LDH漏出量与Cyt-C蛋白表达皆明显上升(P<0.05,P<0.01);与高糖高脂组相比,通心络干预后细胞生存活性、ATP含量和MMP明显升高,细胞上清液LDH漏出量、细胞内ROS水平和Cyt-C蛋白表达明显降低(P<0.05,P<0.01)。结论对于高糖高脂损伤的HAECs,通心络具有保护作用,相关保护机制可能和保护线粒体功能相关。Objective To explore the effect of high glucose and high lipid on human aortic endothelial cells(HAECs)and the intervention effect of Tongxinluo on it.Methods HAECs were randomly divided into the following 5 groups:high,medium and low concentration Tongxinluo groups,high sugar and high lipid group,and blank control group.The cell survival activity,lactic dehydrogenase(LDH)content in cell supernatant,adenosine triphosphate(ATP)content,mitochondrial membrane potential(MMP),intracellular reactive oxygen species(ROS)level and cytochrome(Cyt)-C protein expression of each group were detected.Results Compared with blank control group,the levels of MMP,ATP and cell survival activity were significantly decreased,the ROS content in cells,LDH leakage in cell supernatant and Cyt-C protein expression were significantly increased in high sugar and high lipid group(P<0.05,P<0.01).Compared with the high sugar and high lipid group,the survival activity of cells,ATP content and MMP were significantly increased,while LDH leakage in the supernatant of cells,intracellular ROS level and Cyt-C protein expression were significantly decreased after Tongxinluo intervention(P<0.05,P<0.01).Conclusions For HAECs damaged by high sugar and high lipid,Tongxinluo has a protective effect,and the related protection mechanism might be related to the protection of mitochondrial function.
