摘 要:目的探究米诺环素(Mino)通过调节缺氧诱导因子-1α(HIF-1α)/血管内皮生长因子(VEGF)信号通路对前列腺癌(PCa)细胞增殖、凋亡和血管生成的影响。方法将PC-3细胞分为PC-3组(未做任何处理的PC-3细胞)、L-Mino组(5μM Mino)、M-Mino组(10μM Mino)、H-Mino组(20μM Mino)、H-Mino+二甲基草酰甘氨酸(DMOG)组(20μM Mino、2 mM HIF-1α/VEGF信号通路激活剂DMOG共同孵育PC-3细胞)。检测PC-3细胞增殖、细胞凋亡率、细胞迁移、侵袭、毛细管形成、上皮间质转化(EMT)蛋白及HIF-1α、VEGF蛋白水平。结果与PC-3组相比,L-Mino组、M-Mino组、H-Mino组PC-3细胞生长抑制率、细胞凋亡率、E-钙黏蛋白(E-cadherin)蛋白水平依次显著增加(P<0.05),PC-3细胞划痕愈合率、侵袭数量、毛细管数量、波形蛋白(vimentin)、N-钙黏蛋白(N-cadherin)、HIF-1α、VEGF蛋白水平依次显著下降(P<0.05),呈现剂量相关性;而DMOG消除了这种影响。结论Mino可能通过抑制HIF-1α/VEGF信号通路来抑制PC-3细胞的增殖及血管生成。Objective To investigate the influences of minocycline(Mino)on the proliferation,apoptosis and angiogenesis of prostate cancer(PCa)cells by regulating hypoxia inducible factor-1α(HIF-1α)/vascular endothelial growth factor(VEGF)signaling pathway.Methods PC-3 cells were divided into PC-3 group(PC-3 cells without any treatment),L-Mino group(5μm Mino),M-Mino group(10μm Mino),H-Mino group(20μm Mino),and H-Mino+dimethyloxallyl glycine(DMOG)group(PC-3 cells co incubated with 20μm Mino and 2 mM HIF-1α/VEGF signal pathway activator DMOG).Proliferation,apoptosis rate,cell migration,invasion,capillary formation,the level of epithelial mesenchymal transition(EMT)protein,and HIF-1α,VEGF protein of PC-3 cell were detected.Results Compared with PC-3 group,the growth inhibition rate,apoptosis rate and the level of E-cadherin protein of PC-3 cells in L-Mino group,M-Mino group and H-Mino group were increased obviously in turn(P<0.05),the scratch healing rate,the number of invading cells,the number of capillaries,and the level of vimentin,N-cadherin,HIF-1αand VEGF protein of PC-3 cells were decreased obviously in turn(P<0.05),these indexes showed dose correlation;while DMOG eliminated this effect.Conclusions Mino may inhibit the proliferation and angiogenesis of PC-3 cells by inhibiting HIF-1α/VEGF signaling pathway.
