摘 要:目的:观察枳实生品及麸炒品提取液对脾虚大鼠肝组织能量代谢相关酶活性及对CaMKⅡ通路CaM、CaMKⅡ蛋白表达水平的影响。方法:SD大鼠随机分为7组,空白对照组,利血平模型组,阳性药组,枳实生品高、低剂量组及枳实麸炒品高、低剂量组。除空白对照组外,其余各组均采用利血平注射制成脾虚模型。测定各组大鼠肝匀浆中物质能量代谢相关指标的活性差异,采用蛋白免疫印迹法比较研究肝组织中CaM、CaMKⅡ及p-CaMKⅡ蛋白表达差异。结果:与模型组比较,枳实麸炒品高剂量组钠钾ATP酶(Na+-K+-ATPase)活性升高;枳实麸炒品高、低剂量组钙镁ATP酶(Ca 2+-Mg 2+-ATPase)活性升高;枳实麸炒品高剂量组柠檬酸合成酶(CS)及异柠檬酸脱氢酶(ICD)均升高;枳实生品低剂量组CaM蛋白表达量升高;枳实麸炒品高剂量组CaMKⅡ表达量升高;枳实生制品高、低剂量组p-CaMKⅡ蛋白表达量均升高。结论:枳实麸炒品不论在三磷酸腺苷酶活性或是CaMK II信号通路关键分子蛋白表达方面均较生品有显著提高,说明麸炒品能更好改善脾虚大鼠症状。Objective To investigate the effect of raw and stir-frying with bran Aurantii Fructus Immaturus on the energy metabolizing enzyme activity and expression of CaM and CaMK II protein in CaMK II signaling pathways in the liver tissue of rats with spleen deficiency syndrome.Methods SD rats were randomly divided into 7 groups:normal control group,Reserpine model group,Positive medicine group,Aurantii Fructus Immaturus raw and bran fried high and low dose groups.The spleen deficiency syndrome rat models were generated by reserpine injection,and ATPase activity were evaluated.Western blotting was used to test CaM,CaMK II,p-CaMK II expression in liver tissue of the spleen deficiency syndrome rats.Results Compared with the model group,he activity of Na+-K+-ATPase was increased in the high-dose of stir-frying with bran Aurantii Fructus Immaturus,the activity of Ca 2+-Mg 2+-ATPase was increased in the high and low dose of stir-frying with bran Aurantii Fructus Immaturus.the expression of CS and ICD increased in high-dose of stir-frying with bran Aurantii Fructus Immaturus.The expression of CaM protein was increased in low dose of raw Aurantii Fructus Immaturus,the expression of CaMKⅡincreased in high-dose of stir-frying with bran Aurantii Fructus Immaturus,the expression of p-CaMKⅡincreased in high-dose of raw and stir-frying with bran Aurantii Fructus Immaturus.Conclusion The stir-frying with bran Aurantii Fructus Immaturus could significantly improve the activity of adenosine triphosphatase and the expression of key molecular proteins in CaMK II signaling pathway.It shows that stir-frying with bran Aurantii Fructus Immaturus can better improve the symptoms of deficiency syndrome rats.
