摘 要:制备松香酸(abietic acid,AA)免疫原并对其进行表征。实验采用AA及其衍生物脱氢松香胺(dehydro-rosin amine,DHAM)、12-羟基松香酸(12-hydroxyrosin acid,12-HA)分别与牛血清白蛋白(bovine serum albumin,BSA)偶联制备了3种免疫原,分别与卵清白蛋白(ovalbumin,OVA)偶联制备了3种包被抗原。以基质辅助激光解析飞行时间质谱法(matrix-assisted laser desorption ionization time-of-flight mass spectrometry,MALDI-TOF MS)对免疫原和包被抗原进行表征。结果表明:AA及其衍生物与载体蛋白的偶联率在11~36之间。用3种免疫原分别免疫Balb/c小鼠,获得了多克隆抗血清。其中由AA-BSA作为免疫原免疫小鼠的抗血清效价可达1:6000,并存在一定程度的抑制。AA及其衍生物均已成功与蛋白偶联,其中由AA-BSA作为免疫原免疫小鼠的抗血清可特异性识别AA。本研究为AA单抗的制备及其免疫分析方法的建立奠定了基础。To prepare and characterize the immunogens of abietic acid(AA),AA and its derivatives dehydrorosin amine(DHAM)and 12-hydroxyrosin acid(12-HA)were conjugate to bovine serum albumin(BSA)to obtain three immunogens.To evaluate the anti-bodies,AA,DHAM and 12-HA were also linked to ovalbumin(OVA)to synthesize coating antigens.MALDI-TOF was used to char-acterize the immunogens and coating antigens.The coupling rates of AA,DHAM and 12-HA to carrier protein in the prepared immu-nogens were in range of 1l~36.Balb/c mice were immunized with three immunogens,and the titer of the polyclonal antisera ob-tained from the immunized mice using AA-BSA as the immunogen were found at 1:6000.Among them,the polyclonal antisera ob-tained from the immunized mice using AA-BSA as the immunogen was able to specific recognize AA,and some extent of inhibition was also observed.AA,DHAM and 12-HA were successfully coupled to proteins.This study laid the foundation for the preparation of monoclonal antibody against AA and the establishment of immunological analytical method for the detection of AA.
