摘 要:以红掌新品种“黑皇后”植株的嫩叶、叶柄和根段为外植体,对影响愈伤组织诱导、愈伤组织继代和不定芽分化、再生团块继代和不定芽分化、不定芽生根等因素进行研究,同时采用数学公式推算组培苗的年产量,以期为红掌适度规模化繁育和栽培提供参考依据。结果表明:愈伤组织诱导的最佳外植体是叶片,其次为叶柄,最后为根段,诱导率分别为70%、40%、35%;愈伤组织诱导的最佳培养基为1/2MS+6-BA0.5+2,4-D0.5;愈伤组织继代和不定芽分化的最佳培养基为MS+6-BA1.0+2,4-D0.2;再生团块继代和不定芽分化的最佳培养基为MS+6-BA1.0+NAA0.2;丛生芽生根的最佳培养基为1/2MS+NAA0.2;以芽、嫩茎和根段直接生芽的模式,1瓶原种年产苗量Y=mXn;以再生团块或愈伤组织生芽的模式,1瓶原种年产苗量Y=ac(1-Xn)/(1-X)。The factors affecting the induction of callus tissue,succession of callus tissue and differentiation of adventitious buds,succession of regenerating mass and of adventitious buds,and rooting of adventitious buds were investigated by using young leaves,petioles and root segments of the new variety“Black Queen”as explants,and the annual yield of the seedlings was calculated by using mathematical formula,in order to provide reference for A.andraeanum moderate scale breeding and cultivation.The results showed that the best explants for callus tissue induction were leaves,followed by petioles and finally root segments,with induction rates of 70%,40%and 35%respectively;the best medium for callus tissue induction was 1/2MS+6-BA_(0.5)+2,4-D_(0.5);the best medium for callus tissue succession and adventitious shoot differentiation was MS+6-BA_(1.0)+2,4-D_(0.2).The best medium for the succession and adventitious shoot differentiation was MS+6-BA_(1.0)+NAA_(0.2);the best medium for the rooting of clumped shoots was 1/2MS+NAA_(0.2);the annual seedling yield mathematical formula of one bottle seedlings for the model of direct shoot,shoot and root segment production was Y=mX^(n);the annual seedling yield mathematical formula of one bottle seedlings for the model of regenerating mass or callus tissue for shoot production was Y=ac(1-X^(n))/(1-X).
