摘 要:细菌小RNA(small RNA, sRNA)又称非编码RNA (non-coding RNA, ncRNA),是一组长度为50~500 nt的RNA分子,通常不编码蛋白质,但具有独立的功能。sRNA作为转录后调节因子在细菌的各种生理过程中发挥重要的作用。尽管目前人们已经从各种细菌中鉴定了大量小RNA,但是大多数小RNA的功能还不清楚。本实验室之前在十字花科黑腐病菌(Xanthomonas campestris pv.capestris, Xcc) 8004菌株(简称Xcc 8004)中鉴定了612个小RNA,但绝大多数小RNA的功能仍然未知。本研究采用过表达的方法对小RNA(sRNA-Xcc132)的生物学功能进行鉴定。结果发现,与带有空载质粒的野生型菌株相比,sRNA-Xcc132的过表达菌株OE132在基本培养基中的生长速度明显加快而且对Co^(2+)的耐受力明显增强,说明sRNA-Xcc132与Xcc 8004的生长和Co^(2+)耐受性有关。Bacterial small RNA(small RNA,sRNA),also known as non-coding RNA(non-coding RNA,ncRNA),is a group of RNA molecules that are 50~500 nucleotides in length,normally do not encode proteins,but has independent functions.sRNA acts as a post-transcriptional regulator and plays an important role in various bacterial cellular physiological processes.Although a large num-ber of sRNAs had been identified in dfferent bacterial species,few of them has been functionally characterized.Our laboratory pre-viously identified 612 sRNAs from Xanthomonas campestris pv.campestris strain 8004(Xcc 8004),but the function of most small RNAs is still unknown.In this study,the biological function of the sRNA(sRNA-Xcc132)has been determined by using overexpres-sion method.The results showed that the sRNA-Xcc132 overexpression strain OE132 displayed a significantly increased growth rate in minimal medium and a greatly enhanced tolerance to Co^(2+) compared to the wild-type strain carrying the empty vector plasmid,demon-strating that sRNA-Xcc132 is involved in growth and Co^(2+)tolerance of Xcc 8004.
