摘 要:目的:以ITS2序列和psbA-trnH序列作为DNA条形码,对西藏地区紫堇属植物进行分子鉴定。方法:通过比较两条序列的扩增成功率和测序成功率,分析序列特征,计算种内和种间的遗传距离,构建Neighbor-joining进化树,评价两条序列的鉴定效率。结果:两条序列的扩增成功率和测序成功率均为100%。psbA-trnH序列的最大种内遗传距离均小于最小种间遗传距离,符合DNA条形码对遗传距离的要求。系统进化树分析中,psbA-trnH序列能够有效地区分不同来源的紫堇属植物,而ITS2序列聚类效果较差。结论:psbA-trnH序列具有较高的鉴定效率,可作为西藏地区紫堇属植物物种鉴定的DNA条形码。该研究为西藏地区紫堇属植物的基因组学的深入研究夯实了基础,也为西藏紫堇属藏药的质量监控提供了新的方法。Objective:To use ITS2 sequence andpsbA⁃trnHsequence as DNA barcodes for molecular identification ofCorydalis plants from Tibet.Methods:By comparing the amplification success rate and sequencing success rate of the two sequences,the sequence characteristics were analyzed,the intraspecic and interspecific genetic distances were calculated.Neighbor⁃joining evolutionary tree was constructed to evaluate the identification efficiency of the two sequences.Results:The success rate of amplification and sequencing of the two sequences was 100%.The maximum intraspecific genetic distance ofpsbA⁃trnHsequence was smaller than the minimum inter⁃specific genetic distance,which met the genetic distance requirement of DNA barcodes.In phylogenetic tree analysis,psbA⁃trnHse⁃quence could effectively distinguish differentCorydalisplants,but ITS2 sequence had poor clustering effect.Conclusion:psbA⁃trnHse⁃quence has high identification efficiency and can be used as DNA barcodes for the identification ofCorydalisplants in Tibet.The study lays a solid foundation for the further study on the genomics ofCorydalisplants in Tibet and provides a new method for the quality con⁃trol of Tibetan medicine ofCorydalisplants in Tibet.
