摘 要:目的优选沙苑子总黄酮显色及提取工艺。方法以沙苑子饮片为原料,通过单因素结合正交试验法,研究沙苑子总黄酮的显色方法及最佳显色条件、提取方法及最佳提取工艺。结果以AlCl_(3)法为宜;最佳显色条件为0.8 ml提取液加入1.2 ml 60%乙醇,2%AlCl_(3)0.6 ml、pH 4.5的醋酸-醋酸钠缓冲液2.0 ml,60%乙醇定容,室温静置20 min,于420 nm处测定吸光度;最佳提取工艺为70%乙醇,85℃加热回流提取2 h,料液比1∶25(g/ml),提取2次,得总黄酮的含量为24.5 mg/g。结论该显色和提取方法可行性高,重复性较好,本研究为沙苑子的进一步开发利用提供了实验依据。Objective To optimize the chromogenic and extraction process of total flavonoids from Astragalus complanatus.Methods By using Astragalus complanatus decoction pieces as raw materials,the chromogenic method,optimal chromogenic conditions,extraction method and optimal extraction process of total flavonoids from Astragalus complanatus were studied by single factor combined with orthogonal test.Results AlCl_(3)method was suitable.The optimal chromogenic conditions were as follows:add 1.2 ml of 60%ethanol,0.6 ml of 2%AlCl_(3)and 2.0 ml of pH 4.5 acetic acid-sodium acetate buffer to 0.8 ml of the extract,dilute with 60%ethanol to volume,let it stand at room temperature for 20 min,and measure the absorbance at 420 nm.The optimal extraction process was set as follows:70%ethanol,reflux extraction at 85℃for 2 h,solid-liquid ratio of 1∶25(g/ml),2 extractions in total,and the content of total flavonoids obtained was 24.5 mg/g.Conclusion The chromogenic and extraction method is feasible and reproducible.Therefore,this study provides experimental basis for the further development and utilization of Astragalus complanatus.
