摘  要:目的 探讨静磁场暴露对大鼠抑郁样行为及海马多巴胺受体2(DR2)和多巴胺转运体(DAT)表达的影响。方法 将Wistar大鼠置于自制鼠笼中,采用超导磁体暴露源进行全身暴露,暴露强度为50 mT、100 mT、200 mT,时间为1 h/d,连续暴露15 d或30 d。采用糖水试验、脑电生理检测大鼠在不同强度磁场暴露后行为学及脑电改变,而后采用免疫组化法检测大鼠海马区DR2和DAT的表达并进行定量分析。结果 200 mT静磁场暴露30d致大鼠糖水偏爱指数明显降低;三个暴露组大鼠脑电异常,其中高强度暴露组改变最为显著;免疫组化结果显示,静磁场暴露第10 d, 100 mT和200 mT暴露组海马DAT表达显著升高,暴露第15 d,各暴露组DAT表达均显著升高。暴露第15 d,各暴露组大鼠海马DR2表达均显著降低。结论 静磁场暴露导致大鼠抑郁样行为及脑电改变,海马区DR2和DAT表达异常可能与其抑郁样行为发生相关。Objective To explore the effects of static magnetic field(SMF)exposure on the depression-like behavior of rats and the expression of dopamine receptor 2(DR2)and dopamine transporter(DAT)in rats'hippocampus.Methodss Wistar rats were placed in home-made organic rat cages.Whole-body exposure was carried out using a superconducting magnet source with intensities of 50,100,and 200 mT,for 1 h/d in consecutive 15 or 30 days.Behavioral and electroencephalographic changes in rats induced by SMF with different intensities were detected using the sugar-water test and electroencephalogram detection.Besides that,immunohistochemical SP method was used to detect the expression of DR2 and DAT in the hippocampus of rats,and then quantitative analysis of images was done.Results 200 mT SMF exposure for 30 days resulted in a significant decrease in sugar-water preference index in rats.EEG abnormalities were observed in rats of the three exposure groups,with most significant changes in the 200mT exposure group.Immunohistochemical results showed that the expression of hippocampal DAT was significantly increased in the 100 and 200 mT groups on the 10th day of SMF exposure.On the 15th day of SMF exposure,the expression of DAT in hippocampus was significantly increased in all exposure groups.On the 15th day of exposure,the expression of DR2 was significantly reduced in all groups.Conclusions SMF exposure induced depression-like behavior and EEG changes in rats,and the abnormal expression of DR2 and DAT in the hippocampus might be associated with their depression-like behavior.
