摘 要:目的研究索拉非尼和吴茱萸碱联用对HepG2肝癌细胞增殖的抑制作用及其机制。方法以肝癌HepG2细胞为研究对象,采用CCK-8法检测索拉非尼单独或与吴茱萸碱联用对HepG2细胞增殖抑制率;Annexin V-FITC/PI双染法流式细胞检测细胞凋亡水平;Western blot测定凋亡相关蛋白的表达。结果索拉非尼与吴茱萸碱对HepG2细胞增殖均具有明显抑制作用(P<0.05),两种药物联合应用的细胞凋亡率较单独用药组均明显升高(P<0.05);Western blot结果显示两药联合应用时,Bax、Cleaved caspase-3蛋白表达水平较单独用药组明显增加(P<0.05),Bcl-2蛋白表达水平明显降低(P<0.05)。结论索拉非尼联合吴茱萸碱可通过协同作用显著抑制肝癌HepG2细胞增殖,其机制可能与诱导肿瘤细胞发生凋亡有关。Objective To investigate the inhibitory effect of the combination of sorafenib and evodiamine on the proliferation of HepG2 cells and the underlying mechanism.Methods The inhibitory effects of sorafenib and evodiamine used alone or in combination on the growth of HepG2 cells were evaluated by CCK-8 assay.The apoptosis levels were detected by flow cytometry using the Annexin V-FITC/PI double staining method.The expression changes of apoptosis-related proteins were detected by Western blot.Results Both sorafenib and evodiamine had obvious inhibitory effects on the proliferation of HepG2 cells(P<0.05),and the apoptosis rate of the combined treatment group was significantly higher than that of the single treatment group(P<0.05).Compared with the single use group,the expression levels of Bax and Cleaved caspase-3 proteins in the combination group were significantly increased,while the protein expression level of Bcl-2 was significantly lower than that in the single use group(P<0.05).Conclusion These results suggested that sorafenib and evodiamine had a synergistic antitumor effect on hepatoma HepG2 cells,and the possible mechanism is related to the induction of tumor cell apoptosis.
