摘 要:本研究以牛血清蛋白(BSA)为模板蛋白,在不同条件下合成了BSA@ZIF-8复合物。采用傅里叶红外光谱(FT-IR)、扫描电子显微镜(SEM)、热重分析(TGA)及激光粒度仪对制备的复合物的形貌结构进行了表征分析。结果发现:对比不同锌:二甲咪唑:水的比例下合成的复合物,原料中二甲咪唑的浓度越高,BSA@ZIF-8中BSA负载率和包埋率越低;较高的温度有利于ZIF-8晶体的生成,从而导致较大的复合物粒径和较低的BSA包埋率和负载率。此外,还发现复合物孵育时间需控制在24h左右,超过24h晶体基本不再生长,但BSA包埋率与负载率反而会下降。最后,中性环境(pH=7.4)更有利于BSA的包埋,而酸性或碱性环境则有利于ZIF-8的生长。In this study,BSA@ZIF-8 composites were synthesized under different conditions using bovine serum albumin(BSA)as model protein.The morphology and structure of the composites were characterized and analyzed by Fourier infrared spectroscopy(FT-IR),scanning electron microscopy(SEM),thermo-gravimetric analysis(TGA)and laser particle size analyzer.The results showed that the higher the concentration of dimethylimidazole in the raw material would lead to the lower the loading ratio and embedding ratio of BSA in BSA@ZIF-8 by characterization of the composites obtained under different ratios of zinc,dimethylimidazole and water ratio.A higher temperature was conducive to the formation of ZIF-8 crystals,resulting in larger particle size of composites and lower embedding ratio and loading ratio of BSA.In addition,it was also found that the incubation time of composites should be controlled at around 24 h.After 24 h,the crystal would no longer grow but the embedding ratio and loading ratio of BSA actually decreased.Finally,it was found that neutral environment(pH=7.4)was more conducive to the embedding of BSA,while acid and alkaline environment was beneficial to the growth of ZIF-8.
