摘 要:Fed-batch操作因其能有效改善微生物生长环境、延长发酵时间和消除原料及产物抑制而广泛应用于发酵工业中。从动态优化的理论及数值计算两方面,对发酵过程的最优给料参考轨迹进行分析和模拟计算,结果显示,应用基于有限元的正交配置方法可以较好地处理bang-singular-bang控制问题;当操控输入取在离散点内为零阶保持时,优化的参考轨迹在工业实践中较容易实现,并且具有优于恒定给料的效果。以青霉素发酵过程为例,将优化目标设定为产品/卸料基质浓度时,可获得更优的青霉素产量,并且能有效控制基质的产品转化率,降低后续产品、原料分离成本。该青霉素优化计算方案可推广至一般Fed-batch过程,辅助工业过程建立最优给料策略。Fed-batch operation was widely used in the fermentation industry because it could effectively improve the environment,prolong the reaction time and eliminate the inhibition of substrate and end-products.The optimal feed reference trajectory of penicillin fermentation process was analyzed and numerically presented,and the results showed that the bang-singular-bang control problem could be solved effectively by orthogonal collocation on finite elements,and when the control input took zero-order hold within the finite points,the optimized reference trajectory was easy to implement in practice.As far as the penicillin fermentation process described in this paper was concerned,when the optimization target was set as the concentration of the product/unloading substrate,a better penicillin yield could be obtained,and the product conversion rate could be effectively controlled,and the cost of subsequent product/raw material separation could be reduced.The optimization scheme proposed could be extended to general Fed-batch process to assist industrial process to establish optimal feeding strategy.
