摘 要:以葛花和菊花为原料,以水煎法作为提取方法,采用可见分光光度法和紫外分光光度法对水煎液中总黄酮含量进行测定,从而探索合适的测量葛花菊花水煎液中总黄酮浓度的方法。可见分光光度法以芦丁对照品,采用NaNO_(2)-Al(NO_(3))_(3)-NaOH显色法,紫外分光光度法分别以芦丁和鸢尾苷为对照品,分别进行可见光区和紫外的光谱扫描。结果表明,芦丁对照品溶液显色后在506 nm处有吸收峰位,而样品溶液同条件显色后在可见光范围内无吸收峰位。在紫外分光光度法中,芦丁对照品溶液在258 nm处有最大峰值,而样品溶液在264 nm处有最大吸收峰值,两者的吸收峰位相差6 nm。而鸢尾苷对照品溶液和样品溶液在波长264 nm均有吸收峰,两者吸收峰位一致。以NaNO_(2)-Al(NO_(3))_(3)-NaOH的显色法不适用于测定样品溶液中总黄酮的含量,而以鸢尾苷对照品的紫外分光光度法最适用于葛花菊花水煎液总黄酮含量的测定。通过方法学考察表明该法是一种稳定、精密性和重复性俱佳的方法。该研究为以葛花和菊花为主要原料的解酒药方的开发与应用提供了基础。Puerariae Flos and Chrysanthemum as raw materials,water decoction was used as extraction method.Besides,the content of total flavonoids in water decoction was determined by visible spectrophotometry and ultraviolet spectrophotometry.The objective of this study is to explore a suitable method for measuring the concentration of total flavonoids in the decoction of Puerariae Flos and Chrysanthemum.For visible spectrophotometry,we used Rutin as the control product and used NaNO_(2)-Al(NO_(3))_(3)-NaOH as color development method.For ultraviolet spectrophotometry,we used rutin and irisin as the control product.The sample solution and the control solution were spectroscopically scanned in the visible and ultraviolet range.The result shows that rutin has an absorption peak at 506 nm after color development,while the sample solution has no absorption peak in the visible light range after color development under the same conditions.In the ultraviolet spectrophotometry,the rutin control solution had the maximum absorption peak at 258 nm,while the sample solution had the maximum absorption peak at 264 nm.There is a difference of 6 nm between the absorption peaks.But the control solution and the sample solution have the same absorption peak at 264 nm.Color development method as NaNO_(2)-Al(NO_(3))_(3)-NaOH is not suitable for the determination of total flavonoids and the UV spectrophotometry with irisin as a reference product is the most suitable method for the determination of total flavone content.The method is proved to be stable,precise and accurate by methodological examination.This study provides the basis for the development and application of antialcoholism prescription with Puerariae Flos and Chrysanthemum as main raw materials.
