柑橘黄龙病菌在雷波脐橙中的分布规律及其原噬菌体类型分析

摘　　要：为了解柑橘黄龙病菌在雷波脐橙不同组织部位的含量差异和遗传多样性,为雷波县柑橘黄龙病流行研究提供理论基础,利用Real-time PCR分析黄龙病菌在雷波脐橙8个组织(新梢叶片、新梢韧皮部、果蒂、橘络、果实中轴、枝条叶片、枝条上部韧皮部、枝条下部韧皮部)以及橘络不同分段中的绝对定量,并鉴定其原噬菌体类型。结果表明,黄龙病菌含量在果实组织(果蒂、橘络、中轴)中显著高于新梢组织及枝条组织。黄龙病菌在橘络中含量不均匀,集中分布在果实中间部分。通过原噬菌体类型鉴定发现,新梢组织的病原为Type 2+Type 4类型,而成熟组织的病原为Type 1类型,并且在果蒂中存在两类病原混合感染。综上,黄龙病菌在雷波脐橙中主要富集于果实组织的橘络中部,并且目前雷波县存在外源的黄龙病菌侵染新梢组织的情况。In order to understand the titer difference and genetic diversity of Candidatus Liberibacter asiaticus(CLas)in different tissue of Leibo navel orange,and provide theoretical foundation for the epidemiology of Citrus Huanglongbing in Leibo County,the Real-time PCR was used to analyze the absolute quantitation of CLas in eight tissues(leaf of new shoot,phloem of new shoot,fruit pedicle,fruit pith,fruit central axis,leaf of branch,phloem of upper branch,phloem of lower branch)and different segments of pith of Leibo navel orange,and to identify the prophage type.The results showed that the content of CLas in fruit tissues(pedicle,pith,central axis)was significantly higher than that in shoot tissue and branch tissue.The content of CLas in pith was asymmetrical and concentrated in the middle part of fruit.The prophage type identification showed that CLas was Type 2+Type 4 in the new shoot tissue and Type 1 in the other mature tissue,and there was mixed infection of the two types of CLas in the fruit pedicle.In conclusion,CLas was mainly concentrated in the middle pith of fruit tissue in Leibo navel orange,and at present,there was a situation of external CLas infecting shoot tissue in Leibo County.
