摘 要:本文指出远海梭子蟹是我国重要的经济蟹类之一,具有肉质鲜美、病虫害少、出肉率可观等优点,具有极高的开发价值。本文通过引物设计、PCR扩增、PCR产物测序验证测序结果,准确地获得了远海梭子蟹TTK基因的全长序列。使用生物信息学工具对TTK基因进行分析预测,研究表明TTK基因的CDS序列全长1 752 bp,编码氨基酸583个,其中丝氨酸占比最多,为10.1%,色氨酸占比最少,为0.5%;序列的理论等电点5.19,氨基酸平均亲水性-0.808,是亲水蛋白质;序列中磷酸化位点64个,其中丝氨酸磷酸化位点47个,苏氨酸磷酸化位点17个;蛋白质二级结构中α-螺旋、延伸链、β-折叠、无规则卷曲占比分别为30.87%、7.38%、6.00%、55.75%。远海梭子蟹TTK基因的序列验证与蛋白质分析将为进一步研究该基因的功能奠定基础并提供一定参考。This paper points out that the Portunus pelagicus is one of the important economic crab species in China,with the advantages of delicious meat,low pest and disease,and considerable meat yield,which is of great value for exploitation.The full-length sequence of TTK gene of Portunus pelagicus is accurately obtained by primer design,PCR amplification and PCR product sequencing to verify the sequencing results.Using bioinformatics tools to analyse and predict the TTK gene,the study showed that the CDS sequence of TTK gene is 1752 bp in length,encoding 583 amino acids,of which serine accounts for the most,10.1%,and tryptophan accounts for the least,0.5%;the theoretical isoelectric point of the sequence is 5.19,and the average hydrophilicity of amino acids is-0.808,which is a hydrophilic protein;the sequence has 64 phosphorylation sites,including 47 serine phosphorylation sites and 17 threonine phosphorylation sites;the percentages of alpha-helix,extended chain,beta-fold and irregular curl in the secondary structure of the protein are 30.87%,7.38%,6.00%and 55.75%respectively.The sequence validation and protein analysis of the TTK gene of Portunus pelagicus will provide a basis and reference for further study of the function of this gene.
