摘 要:为有效解决湛江地区甘薯病毒病影响甘薯品质与产量的问题,采用快速催芽与植物茎尖组织培养技术结合的方式快速繁殖甘薯;运用微茎尖分生组织技术获得甘薯无病毒植株并扩繁;利用试管内靠接、研磨液浸染、无糖培养嫁接、水培嫁接、分子生物学检测等5种方式进行病毒检测,获得无病毒植株和有效的甘薯病毒检测方法。试验结果表明:通过热处理结合微茎尖分生组织技术可有效减少甘薯携带病毒种类,实现甘薯种苗脱毒;试管内靠接、无糖培养嫁接、水培嫁接等3种嫁接方式和分子生物学检测均可有效检测甘薯病毒。To effectively solve the problem of sweet potato virus disease affecting the quality and yield of sweet potato in the Zhanjiang area,this study adopted the rapid propagation of sweet potato by combining rapid germination and plant stem tip tissue culture technology,used micro stem tip meristematic tissue technology to obtain virus-free plants of sweet potato and propagate them;and utilized five methods of virus detection,including leaning grafting in a test tube,milling solution dipping,grafting with unsweetened culture,grafting by hydroponics and molecular biology detection to obtain virus-free plants and effective sweet potato virus detection methods.The experimental results showed that heat treatment combined with micro stem tip meristematic tissue technology could effectively reduce the number of virus species carried by sweet potato and realize the detoxification of sweet potato seedlings;the three grafting methods of in vitro lean grafting,sugar-free culture grafting,hydroponic grafting and molecular biology detection could effectively detect sweet potato viruses.
